Molecular mimicry and functional convergence of bacterial proteins in host-microbe interactions.

Bacterial proteins modulate host immunity through diverse molecular strategies that are often broadly described as molecular mimicry. However, inconsistent terminology has obscured distinctions between genuine host-like interactions and other forms of host-directed immune modulation. We propose an operational framework that classifies bacterial proteins into four mechanistic categories: molecular mimicry, mimic-like behavior, functional convergence, and pathway interference. The framework prioritizes explicit criteria, including host-target engagement, interaction-interface similarity, functio

Linked help from bacterial proteins drives autoantibody production in small vessel vasculitis.

The small vessel vasculitides granulomatosis with polyangiitis (GPA) and microscopic polyangiitis are associated with autoantibodies to neutrophil cytoplasm antigens (ANCA), principally proteinase-3 (PR3) and myeloperoxidase (MPO). There is an association between GPA and nasal carriage of Staphylococcus aureus. The recent finding that S. aureus produces proteins that bind tightly to and block the function of both PR3 and MPO suggests a mechanism for ANCA formation. The bacterial protein-autoantigen conjugate is recognised by B cells with ANCA specificity, internalised, and the bacterial protei

Homologies of the thyroid sodium-iodide symporter with bacterial and viral proteins.

We have demonstrated that Na+/I- symporter (NIS), a novel thyroid autoantigen, has local amino acid sequence homologies with the other thyroid autoantigens: Thyroglobulin (Tg), thyroid peroxidase (TPO) and thyrotropin receptor (TSH-R). These homologies concern the 4th, 5th, 6th extracellular loop and the beginning of the intracellular tail. We have expanded our studies and found that there are significant local homologies with other 11 proteins, most of them of bacterial or viral origin (e.g., Streptococcus or Herpes). These homologies concern the 2nd and 4th extracellular loop, and both the b

Interaction mechanisms of MXene with bacterial division proteins: towards efficient antibacterial materials.

CONTEXT: The rising threat of bacterial infections, coupled with the rise of antibiotic-resistant strains, has driven the search for alternative antibacterial strategies that do not rely on traditional chemical agents. Among these, antibacterial materials, particularly those based on two-dimensional nanomaterials, have emerged as promising platforms for preventing bacterial adhesion and proliferation. In this context, MXenes, a class of transition metal carbides and nitrides, have recently gained attention for their potent antibacterial properties. However, the molecular-level mechanisms by wh

UbiB proteins mediate an ATP-dependent decarboxylation step in bacterial ubiquinone biosynthesis.

Polyisoprenoid quinones such as ubiquinone (UQ) play an essential role in cellular physiology, acting as membrane-bound electron and proton carriers in respiratory chains and other biological processes across all domains of life. In Escherichia coli, the canonical UQ biosynthesis pathway is well characterized. It involves 12 proteins (UbiA-UbiK and UbiX), most of which catalyze one of the eight modifications of the aromatic ring derived from 4-hydroxybenzoic acid (4-HB), while others (UbiB, UbiJ, UbiK) act as accessory factors ensuring efficient UQ production. Following prenylation by UbiA and

Purification of Bacterial Lipopolysaccharide, and How to Study Its Interactions with Proteins.

Lipopolysaccharides (LPS) are an important component of the outer membrane of Gram-negative bacteria. Although LPS is not homogeneous and it varies among bacterial species and even from strain to strain, there are significantly conserved elements that confer and dictate the physicochemical properties of this molecule. Isolating and purifying LPS from its source organism provides the means to investigate the interactions of LPS molecules with other binding partners, such as proteins. Here, we describe methods to isolate LPS and discuss methods for interaction studies.

PRRSV promotes bacterial infection by remodeling actin cytoskeleton and cell membrane proteins.

UNLABELLED: Secondary infection is a worldwide problem in the prevention and control of viral infection. Secondary bacterial infection induced by porcine reproductive and respiratory syndrome virus (PRRSV) infection causes enormous economic losses, but the relevant mechanism remains unclear. We found that the infection of Klebsiella pneumoniae or Streptococcus suis type 2 in the lungs of PRRSV-challenged piglets was significantly higher than the controls, and the infection of PRRSV, influenza A virus H1N1 (H1N1), and porcine circovirus type 2 (PCV2) also significantly increased the infection o

The Virulent Hypothetical Proteins: The Potential Drug Target Involved in Bacterial Pathogenesis.

Hypothetical proteins (HPs) are non-predicted sequences that are identified only by open reading frames in sequenced genomes, but their protein products remain uncharacterized by any experimental means. The genome of every species consists of HPs that are involved in various cellular processes and signaling pathways. Annotation of HPs is important as they play a key role in disease mechanisms, drug designing, vaccine production, antibiotic production, and host adaptation. In the case of bacteria, 25-50% of the genome comprises HPs, which are involved in metabolic pathways and pathogenesis. The

Meat, dairy and plant proteins alter bacterial composition of rat gut bacteria.

Long-term consumption of red meat has been considered a potential risk to gut health, but this is based on clinic investigations, excessive intake of fat, heme and some injurious compounds formed during cooking or additions to processed meat products. Whether intake of red meat protein affects gut bacteria and the health of the host remains unclear. In this work, we compared the composition of gut bacteria in the caecum, by sequencing the V4-V5 region of 16S ribosomal RNA gene, obtained from rats fed with proteins from red meat (beef and pork), white meat (chicken and fish) and other sources (

Lactoferrin binding protein B - a bi-functional bacterial receptor protein.

Lactoferrin binding protein B (LbpB) is a bi-lobed outer membrane-bound lipoprotein that comprises part of the lactoferrin (Lf) receptor complex in Neisseria meningitidis and other Gram-negative pathogens. Recent studies have demonstrated that LbpB plays a role in protecting the bacteria from cationic antimicrobial peptides due to large regions rich in anionic residues in the C-terminal lobe. Relative to its homolog, transferrin-binding protein B (TbpB), there currently is little evidence for its role in iron acquisition and relatively little structural and biophysical information on its inter

Role of a highly conserved bacterial protein in outer membrane protein assembly.

After transport across the cytoplasmic membrane, bacterial outer membrane proteins are assembled into the outer membrane. Meningococcal Omp85 is a highly conserved protein in Gram-negative bacteria, and its homolog Toc75 is a component of the chloroplast protein-import machinery. Omp85 appeared to be essential for viability, and unassembled forms of various outer membrane proteins accumulated upon Omp85 depletion. Immunofluorescence microscopy revealed decreased surface exposure of outer membrane proteins, which was particularly apparent at the cell-division planes. Thus, Omp85 is likely to pl

Novel role of surfactant protein A in bacterial sinusitis.

BACKGROUND: Chronic rhinosinusitis (CRS) is a common inflammatory disorder of the upper airway characterized by chronic inflammation and significant sinonasal remodeling. CRS is comprised of 2 major subgroups, based on whether polyps are present or absent. In some cases, it is characterized by colonization with opportunistic pathogens such as Pseudomonas aeruginosa (PA), Staphylococcus aureus, and other bacteria. The innate immune system of the sinonasal epithelium is the first line of defense against inhaled pathogens. Surfactant protein A (SP-A) is a member of the collectin family secreted b

Roles of heparin-binding protein in bacterial infections.

Infectious diseases remain a major health problem, where sepsis and other severe infectious diseases are common causes of morbidity and mortality. The importance of early and appropriate treatment of sepsis and severe bacterial infections has been underlined by the successes of measures like the Surviving Sepsis Campaign, among others. Thus, there is a need for clinical and laboratory tools to identify a patient with severe infection early and to distinguish between bacterial and non-bacterial conditions. Heparin-binding protein (HBP) is also called azurocidin, or cationic antimicrobial protei

Functionalizing bacterial cell surfaces with a phage protein.

Functionalization of bacterial cell surfaces has the potential to introduce new activities by chemical modification. Here we show that a bacteriophage-receptor complex can be used to functionalize the surface of two Gram-negative proteobacteria, Escherichia coli and Ralstonia eutropha with CdSe/ZnS nanoparticles. This work highlights the potential for using microbe-phage interactions to generate new functions on living cells.

Synthetically designed anti-defense proteins overcome barriers to bacterial transformation and phage infection.

Bacterial defense systems present considerable barriers to both phage infection and plasmid transformation. These systems target mobile genetic elements, limiting the efficacy of bacteriophage-based therapies and restricting genetic engineering applications. Here, we employ a de-novo protein design approach to generate proteins that bind and inhibit bacterial defense systems. We show that our synthetically designed proteins block defense, and that phages engineered to encode the synthetic proteins can replicate in cells that express the respective defense system. We further demonstrate that a

Phytomodulatory latex proteins improve blood homeostasis in diabetic mice with disseminated bacterial infection.

Blood rheology and hematology are essential factors in vascular health in diabetic patients. Additionally, in patients with systemic infections, blood characteristics change rapidly, increasing cardiovascular risk. The present study examined the role of immunomodulatory proteins from Calotropis procera latex (CpLP) on blood homeostasis in diabetic mice with disseminated bacterial infection. A virulent strain of Salmonella was used as the infection model. Employing the Bingham fluid model, it was determined that CpLP therapy diminished blood yield stress by 75% and decreased the flow behavior i

Acetylated bacterial proteins as potent antigens inducing an anti-modified protein antibody response.

OBJECTIVE: Gut-residing bacteria, such as Escherichia coli, can acetylate their proteome under conditions of amine starvation. It is postulated that the (gut) microbiome is involved in the breach of immune tolerance to modified self-proteins leading to the anti-modified protein antibodies (AMPAs), hallmarking seropositive rheumatoid arthritis (RA). Our aim was to determine whether acetylated bacterial proteins can induce AMPA responses cross-reactive to modified self-proteins and be recognised by human AMPA (hAMPA). METHODS: E. coli bacteria were grown under amine starvation to generate endog

Role of CRISPR-Cas systems and anti-CRISPR proteins in bacterial antibiotic resistance.

The emergence and development of antibiotic resistance in bacteria is a serious threat to global public health. Antibiotic resistance genes (ARGs) are often located on mobile genetic elements (MGEs). They can be transferred among bacteria by horizontal gene transfer (HGT), leading to the spread of drug-resistant strains and antibiotic treatment failure. CRISPR (clustered regularly interspaced short palindromic repeats)-Cas (CRISPR-associated genes) is one of the many strategies bacteria have developed under long-term selection pressure to restrict the HGT. CRISPR-Cas systems exist in about hal

Activation of Neutrophils via IP3 Pathway Following Exposure to Demodex-Associated Bacterial Proteins.

Rosacea is a chronic inflammatory condition that predominantly affects the skin of the face. Sera from rosacea patients display elevated reactivity to proteins from a bacterium (Bacillus oleronius) originally isolated from a Demodex mite from a rosacea patient suggesting a possible role for bacteria in the induction and persistence of this condition. This work investigated the ability of B. oleronius proteins to activate neutrophils and demonstrated activation via the IP3 pathway. Activated neutrophils displayed increased levels of IP1 production, F-actin formation, chemotaxis, and production

Binary bacterial toxins: biochemistry, biology, and applications of common Clostridium and Bacillus proteins.

Certain pathogenic species of Bacillus and Clostridium have developed unique methods for intoxicating cells that employ the classic enzymatic "A-B" paradigm for protein toxins. The binary toxins produced by B. anthracis, B. cereus, C. botulinum, C. difficile, C. perfringens, and C. spiroforme consist of components not physically associated in solution that are linked to various diseases in humans, animals, or insects. The "B" components are synthesized as precursors that are subsequently activated by serine-type proteases on the targeted cell surface and/or in solution. Following release of a
