Cell mixing in the spinal cords of mouse chimeras.

With the aim of determining whether there is significant cell mixing during development of the spinal cord, experimental chimeric mice containing two genetically distinct cell populations were produced by aggregating BALB/c or BALB/c x LPT hybrid embryos with C3H/HeN embryos. The BALB/c and LPT hybrid spinal cord cells were distinguished histochemically from the C3H/HeN spinal cord cells by using beta-glucuronidase as an independent cell genotype marker. BALB/c and LPT hybrid cells have high levels of beta-glucuronidase activity, while the C3H/HeN cells have low levels. The spinal cords of the

Infectious enveloped RNA virus antigenic chimeras.

Random insertion mutagenesis has been used to construct infectious Sindbis virus structural protein chimeras containing a neutralization epitope from a heterologous virus, Rift Valley fever virus. Insertion sites, permissive for recovery of chimeric viruses with growth properties similar to the parental virus, were found in the virion E2 glycoprotein and the secreted E3 glycoprotein. For the E2 chimeras, the epitope was expressed on the virion surface and stimulated a partially protective immune response to Rift Valley fever virus infection in vivo. Besides providing a possible approach for de

A plant vesicle-dendritic cell chimera for enhancing cancer immunotherapy.

The tumour microenvironment (TME) causes mitochondrial dysfunction in resident dendritic cells (DCs), resulting in inadequate antigen presentation and weak T cell priming. Herein, we identify hypoxia as a key factor for causing pathological mitochondrial fission in tumour-associated DCs, and develop a plant vesicle-DC chimera to relieve hypoxia-induced mitochondrial dysfunction for enhancing cancer immunotherapy. The biohybrid chimera is fabricated by loading algae-derived nanovesicles (ANVs) with genetically engineered CCR2 overexpressing-DCs. The CCR2-DC-ANVs target tumour by leveraging the

Autoimmunity and hyperactivation of immune responses by HAE-FLS2 chimera.

Plants employ cell surface receptors to perceive extracellular signals and initiate appropriate cellular responses, thereby regulating diverse physiological processes. The well-characterized leucine-rich repeat receptor-like kinases (LRR-RLK) type receptor FLAGELLIN-SENSITIVE 2 (FLS2) functions as a pattern recognition receptor (PRR) that specifically detects bacterial flagellin, activating downstream responses including MAPK signaling and ROS burst. Here, we report that engineered expression of a chimeric receptor combining the N-terminal extracellular and transmembrane domains of HAE with th

Internally Fabricated Chimera Anterolateral Thigh-Amputate Flaps for Metachronous Reconstruction.

UNLABELLED: The chimera flap is a versatile and exciting tissue composition for the reconstruction of complex tissue defects. METHODS: Here, we present 2 cases of internally fabricated anterolateral thigh (ALT)-amputate chimera flaps for metachronous transfer. RESULTS: A 22-year-old man (case 1) developed extended soft tissue necrosis in both legs following meningococcal septicemia. Before unilateral amputation, a fasciocutaneous flap based on the posterior tibial artery perforators was saved and ectopically implanted onto the ipsilateral thigh, creating an ALT-amputate chimera flap. Chasing dizzy chimera: Diagnosis of combined peripheral and central vestibulopathy.

Diagnosis of combined peripheral and central vestibulopathy remains a challenge since the findings from peripheral vestibular involvements may overshadow those from central vestibular disorders or vice versa. The aim of this study was to enhance detection of these intriguing disorders by characterizing the clinical features and underlying etiologies. We had recruited 55 patients with combined peripheral and central vestibulopathy at the Dizziness Clinic of Seoul National University Bundang Hospital from 2003 to 2013. Peripheral vestibular involvement was determined by decreased caloric respons

Synthetic self-assembling clostridial chimera for modulation of sensory functions.

Clostridial neurotoxins reversibly block neuronal communication for weeks and months. While these proteolytic neurotoxins hold great promise for clinical applications and the investigation of brain function, their paralytic activity at neuromuscular junctions is a stumbling block. To redirect the clostridial activity to neuronal populations other than motor neurons, we used a new self-assembling method to combine the botulinum type A protease with the tetanus binding domain, which natively targets central neurons. The two parts were produced separately and then assembled in a site-specific way

A chimera antibody erythroimmunoassay for detecting HBsAg in human sera.

A highly efficient chimera antibody, a monoclonal anti-hepatitis-B-surface (anti-HBs) antibody coupled with polyclonal anti-sheep-red-blood-cell (anti-SRBC) antibody was prepared using a heterobifunctional reagent, N-succinimidyl-3-(2-pyridyldithiopropionate) (SPDP). Using SRBC as a marker, we established a sensitive solid-phase chimera antibody erythroimmunoassay (CAEIA) according to Guesdon's method. The sensitivity of this assay was 2-20 times higher than the reverse passive haemagglutination assay (RPHA) for detecting HBsAg in serially diluted sera from 10 hepatitis B patients. The weakest

Hybrids/Conjugates/Chimera Drugs-Antimicrobial Hybrids: Antibiotics, Antifungals, Antituberculars, Antimalarials.

Antimicrobial hybrids are compounds that can inhibit, stop the growth of, or kill microorganisms, including bacteria, fungi, and parasites. Antibiotics, a subset of antimicrobial agents, specifically target bacteria and include well-established classes such as β-lactams, macrolides, quinolones, and oxazolidinones. Other antimicrobial hybrids are designed for treating a wide range of diseases, including fungal infections, leishmaniasis, parasitic diseases (such as trypanosomiasis and malaria), leprosy, and tuberculosis. Some hybrids are designed to treat a variety of diseases. This review highl

Systematic mapping of chimera-permissive sites by CRISPR-guided PAM scanning.

Protein chimeras enable new biological functions by combining otherwise independent protein elements, but identifying insertion sites that preserve host protein function remains largely empirical. Here, we introduce CRISPR-guided protospacer adjacent motif (PAM) scanning, a scalable strategy for mapping chimera-permissive sites directly in living cells. PAM scanning uses CRISPR-addressable positions across coding sequences to systematically install functional payloads and measure insertion tolerance. In yeast, PAM accessibility provided near-comprehensive coverage across proteins of diverse si

Systematic mapping of chimera-permissive sites by CRISPR-guided PAM scanning.

Engineered protein chimeras enable new biological functions but remain difficult to design due to context-dependent constraints on insertion tolerance and the need to preserve host protein function. Here, we report CRISPR-guided protospacer adjacent motif (PAM) scanning in yeast to map chimera-permissive sites in living cells. We apply this approach to peptide and reporter insertions. In the first application, we generated 91 insertion chimeras encoding a defined protease cleavage sequence across six components of a model G protein-coupled receptor (GPCR) signaling pathway. Sixty-three percent

Evolutionary Kuramoto dynamics unravels origins of chimera states in neural populations.

Neural synchronization is central to cognition. However, incomplete synchronization often produces chimera states, where coherent and incoherent dynamics coexist. Recent studies have suggested that these chimera states could be important in human cognitive organization. In particular, chimera states have been suggested as a regulator of cognitive integration and regulation with varying quality as humans age. While previous studies have explored such chimera states using networks of coupled oscillators, it remains unclear why neurons commit to communication or how chimera states persist. Here,

Staphylococcus aureus sequence type 71 is a chimera that emerged twice.

BACKGROUND: Staphylococcus aureus is a Gram-positive coccus that colonises or infects humans and a wide range of domestic and wild animals. Various lineages of S. aureus are adapted to ruminants, including cattle, where they commonly cause mastitis. This is of significant animal welfare and economic relevance. Clonal complex (CC) 97 is one of the S. aureus lineages found predominantly in bovines, but also in other animals and humans. Sequence Type (ST) 71 is a related bovine-associated lineage with part of its genome identical to CC97. Thus, it has been assumed that ST71 was a chimera consisti

[Targeting chimera technology: A new tool for undruggable in breast cancer].

Breast cancer is one of the most common and fatal malignancies among women worldwide, and its treatment efficacy is often limited by drug resistance and the presence of undruggable targets. Traditional small-molecule drugs have difficulty effectively modulating certain critical targets such as transcription factors and non-coding RNAs, necessitating new therapeutic strategies. Proteolysis-targeting chimeras (PROTACs) function by recruiting pathogenic proteins to the cellular ubiquitin-proteasome system, thereby inducing their specific degradation. In contrast, ribonuclease-targeting chimeras (

A human chimera for von Willebrand disease following bone marrow transplantation.

PURPOSE: von Willebrand disease (vWD) is a common bleeding disorder characterized by quantitative or qualitative defects in von Willebrand factor (vWF), a protein important for coagulation and platelet adhesion. There are two forms of vWF currently recognized: plasma vWF synthesized by endothelial cells and platelet vWF produced within megakaryocytes. Although both plasma and platelet vWF play important roles in overall hemostasis, the relative contribution of each form is not clear. PATIENTS AND METHODS: We report the results of bone marrow transplantation from a donor with Type I vWD into a

Identification and characterization of TrxA: a novel bacterial thioredoxin-metallothionein chimera.

Heavy metals (HMs) are naturally occurring elements which can be essential, such as zinc, copper, and iron, or non-essential, including cadmium, mercury, and lead. While essential metals serve as cofactors in critical enzymatic processes, elevated concentrations of both essential and non-essential HMs pose severe toxicity risks, primarily through oxidative stress, disruption of metal homeostasis, and biomolecular damage. Microorganisms have evolved diverse mechanisms to cope with metal-induced stress, including metal sequestration, enzymatic transformation, efflux systems, and surface immobili

Development of Oncolytic Virus-Proteolysis-Targeting Chimera Conjugates for Tumor Immunotherapy.

The development of proteolysis-targeting chimeras (PROTACs) represents a promising strategy for targeted protein degradation in cancer therapy. However, the limited tumor-specific targeting and the inherent unfavorable physicochemical properties of PROTACs lead to insufficient cellular uptake and suboptimal antitumor immune responses. Herein, as a proof of concept, we developed an oncolytic virus-PROTAC conjugate (BPAD) by efficiently coupling bromodomain-containing protein 4 (BRD4)-targeting PROTACs with oncolytic viruses (OVs). In BPAD, the potent and highly selective infection of OVs to tum
