Flexibility of cell fates and functions across sex determination systems revealed by comparative single-cell analyses.

Sex determination in vertebrates can be initiated by a wide range of genetic or environmental triggers. Yet, the degree to which gonadal cell types and genetic programs are conserved remains unresolved. Here we employed single-cell transcriptomics to characterize the temperature-dependent sex determination (TSD) program in gonads from the turtle species Trachemys scripta. Comparative analyses against species with genetic sex determination, like mouse (XY) and chicken (ZW), revealed a marked divergence in cell type repertoires and functions during vertebrate evolution. Unlike mammals, fetal Ley

Family Experiences Surrounding Gender Identity Disclosure in TGNC Youth (A Scoping Review).

In recent years, the disclosure of transgender and gender non-conforming (TGNC) individuals identity has emerged as a phenomenon receiving increasing attention from researchers, due to its impact on family dynamics and the mental health outcomes of this population. This research aims to map existing knowledge regarding the experiences of parents of TGNC youth during the gender identity disclosure process, between 2018 and 2024. To conduct this research, the Joanna Briggs Institute (JBI) methodology for scoping reviews and the PRISMA guidelines were used. Five major themes emerged from this pro

Spatiotemporal transcriptome atlas of human embryos after gastrulation.

The comprehensive spatiotemporal atlas of gene expression during early human embryonic development is critical for insights into embryogenesis1, organogenesis2 and disease origins3,4. Here, leveraging Stereo-seq technology, we generated spatial transcriptomic profiles across 77 sagittal sections of 13 whole-human embryos ranging from Carnegie stage 12 to 23, integrated with single-nucleus RNA sequencing to elucidate gene expression patterns within defined cellular contexts, revealing the cellular heterogeneity that drives organ-specific differentiation. Our study has established a regulatory p

Integrated Breeding Approaches for Ascochyta Blight Resistance in Chickpea.

Ascochyta blight (AB), caused by the necrotrophic fungus [Ascochyta rabiei (Pass.) Labr.], is one of the most destructive diseases of chickpea (Cicer arietinum L.), causing yield losses of up to 100% under favorable conditions. The pathogen possesses a heterothallic mating system with two mating-type idiomorphs, MAT1-1 and MAT1-2, which contribute to high genetic diversity and the frequent breakdown of host resistance. This review summarizes current knowledge of AB biology, epidemiology, and management, highlighting recent advances in molecular diagnostics, host-pathogen interactions, and popu

Energy Transfer Pathways in Plant Photosystem I from First-Principles Modeling.

The photosynthetic supercomplex Photosystem I-light harvesting complex I (PSI-LHCI) of plants is a molecular machine involved in energy conversion. Concentration of energy in the PSI reaction center is strikingly efficient, despite the unusual presence of low-energy, trapping states (red forms) spatially far from the reaction center. The main energy transfer processes occurring in PSI-LHCI are still unclear, as the size and complexity of the system prevent immediate interpretation of spectroscopic signals and challenges traditional modeling approaches. Here we present a multiscale quantum chem

Rapid robust high-fidelity 3D neuronal extraction from multiview calcium imaging datasets.

Recent developments in imaging facilitate large-scale three-dimensional (3D) neuronal recording. While the resulting large datasets shed light on population-level neural coding, extracting neuronal calcium dynamics from 3D volumes remains more challenging than from two-dimensional images due to noise and scattering. Here we present DeepWonder3D, a general end-to-end pipeline for rapid and robust 3D neuronal extraction with high fidelity. Instead of processing voxel by voxel, DeepWonder3D works on the multiview projections of 3D imaging data obtained either digitally or optically through specif

Wild genes to the rescue: high-throughput genomics reveals the wild source of broomrape resistance in sunflower.

The co-evolutionary arms race between crops and their parasites requires continuous identification of new resistance mechanisms. Broomrape (Orobanche cumana), a root parasitic plant, poses a severe threat to sunflower (Helianthus annuus) production, yet the genetic architecture underlying host resistance remains poorly understood. To address this, we established a high-throughput phenotyping platform to quantify root infestation across a diverse sunflower association mapping (SAM) population. Combining this phenotypic resource with a dual genome-wide association study (GWAS) strategy based on

Whole-genome sequence of a female Loa loa adult worm from Cameroon.

OBJECTIVE: Although loiasis affects more than 20 million people and that its associated clinical complications can cause multiple organ failures, this disease remains understudied. Consequently, the elimination of loiasis is not on the agenda of public health services. Defining an effective strategy to tackle this disease is challenging and requires a better understanding of the parasite's biology, including its genetic aspects. Loa loa adult worms provide large amounts of biological tissues for genetic sequencing, but are very difficult to collect. Therefore, limited genomic data are availabl

A novel mathematical modeling and optimal control analysis of monkeypox transmission incorporating double-dose vaccination strategies: Insights from the recent outbreak.

Monkeypox (Mpox) has re-emerged as a serious global public health concern due to its potential human-to-human transmission and persistence in the environment. This study develops a new deterministic model incorporating double-dose vaccination approach to analyze the transmission dynamics and effective interventions for the Mpox outbreak. The model divides the human population in six groups and includes an environmental reservoir, capturing both direct transmission from infectious individuals and indirect transmission through environmental contamination. Global stability results of the equilibr

Revisiting Animal Reservoirs of Hantavirus and Other Zoonotic Viruses Associated with Cruise Travel: Impact and Prevention.

The sustained growth of international maritime tourism and the expansion of the cruise industry have increased the importance of emerging zoonotic diseases as a significant challenge to global public health. Cruise ships constitute complex epidemiological settings characterized by high population density, rapid international mobility, and constant interaction between humans, animals, and coastal environments, factors that favor the introduction and spread of emerging infectious agents. Potential zoonotic concerns include orthohantaviruses and other zoonotic viruses, such as dengue, chikungunya

Thermal physiology of dung beetles exposed to ivermectin, a veterinary drug.

Global changes, including increasing temperatures and pesticide contamination threaten insect survival and reproduction by altering metabolism and stress responses. Of particular importance are insects that provide ecosystem services and are threatened by multiple stressors, such as dung beetles, which bury dung in forests and cattle pastures. This study investigated how elevated temperature and ivermectin, a common antiparasitic medication used in cattle that is excreted in dung, affect the thermal physiology of Euoniticellus intermedius dung beetles under controlled laboratory conditions. Impact of fluoride exposure on reproductive health: Insights into molecular mechanisms and health implications.

While the benefits of fluoride in preventing dental caries are well-established, concerns about its potential toxicity at high intake levels are rising. This review investigates the link between chronic fluoride exposure and reproductive health outcomes at the molecular level, focusing on population growth and child sex ratios in the fluorosis-affected and non-fluorosis regions. The exploration of the detrimental effects of fluoride on both male and female reproductive systems is necessary. In males, hormonal variations, alterations in spermatogenesis, capacitation, and sperm motility using mo

A Qualitative Study Exploring How the Perspectives and Experiences of Cisgender Black Women Inform Their Readiness to Consider Pre-Exposure Prophylaxis for HIV Prevention.

Attention to increasing pre-exposure prophylaxis (PrEP) use among cisgender Black women (CBW) in the southern United States (U.S.) is necessary to achieve national 2030 Ending the HIV Epidemic (EHE) goals. Qualitative exploration of CBW's readiness to use PrEP is necessary to discern whether practical solutions to addressing PrEP uptake within this HIV-vulnerable population are feasible. Focus group discussions (n = 5) and key informant interviews (n = 3) in two EHE jurisdictions in Houston and Austin, Texas were used to explore how perspectives and lived experiences may serve as facilitators

Copy number flexibility facilitates heteroresistance to increasing antibiotic pressure and threatens the beta-lactam pipeline.

Our understanding of the rapid adaptation of bacteria to resist novel drugs is growing beyond known mechanisms such as mobile genetic elements and mutation selection. Heteroresistance (HR) is a form of antibiotic resistance where a phenotypically unstable minority resistant subpopulation co-exists with a susceptible population. We sought to uncover the mechanism of heteroresistance to cefiderocol, a novel β-lactam developed to resist β-lactamases including extended-spectrum-β-lactamases (ESBLs), which has been recently reported but poorly understood. We observe HR to cefiderocol among clinical

The role of platelet transfusion in modulating platelet indices and leukocyte dynamics in dengue fever patients.

Dengue fever, a viral illness transmitted by Aedes mosquitoes, is marked by a reduction in platelet count (PLT) and changes in blood parameters, which increase the risk of complications such as hemorrhage. Platelet transfusion is frequently used to counter thrombocytopenia in dengue cases, but the precise effects on platelet indices (such as mean platelet volume [MPV] and platelet distribution width [PDW]) and leukocyte counts are still not well defined. This literature review investigates the role of platelet transfusions in modifying platelet and leukocyte dynamics in dengue patients. Studie
