ROS produced in mitochondria entrapped by self-assembly peptide fibers for target therapy of glioma.

Brain glioma is a highly energy-dependent malignant tumor. Sonodynamic therapy (SDT) provides a noninvasive and effective approach for brain glioma therapy. Reactive oxygen species (ROS) from sonosensitizers in the treatment of SDT play a key role. Inspired by spider webs, a self-assembling "spider peptide" (P1) bearing porphyrin moieties was constructed to generate ROS under ultrasound. In glioma cells, P1 forms web-like nanofibers that weave around mitochondria and enables ROS to release in situ. This efficiently disrupts the energy metabolism of mitochondria leading to the inhibition of gli

Focused ultrasound-mediated nanocarrier delivery across the blood-brain barrier for neurodegenerative diseases.

The development of effective therapies for neurodegenerative disorders such as Alzheimer's disease, Parkinson's disease, Huntington's disease, and amyotrophic lateral sclerosis remains a major challenge due to the restrictive nature of the blood-brain barrier (BBB). Conventional systemic drug delivery strategies often fail to achieve sufficient central nervous system (CNS) penetration while avoiding peripheral toxicity. Focused ultrasound (FUS), particularly when combined with microbubbles or nanocarriers, has emerged as a non-invasive approach to transiently and precisely open the BBB, enabli

Lactylation Signatures as Predictors of Prognosis and Therapeutic Response in Lung Adenocarcinoma: Implications for Ultrasound-Based Therapeutic Strategies.

BACKGROUND: Lactylation, a unique post-translational alteration, has been identified as an important epigenetic regulator of cancer metabolism, immune evasion, and treatment resistance. Recent research reveals that ultrasound-based biophysical stimulation may change tumor microenvironmental variables that affect lactate metabolism and subsequent lactylation processes. The purpose of this study was to better understand the predictive significance of lactylation-associated genes in lung adenocarcinoma (LUAD) and how they could interact with ultrasound-mediated treatment response. METHODS: Trans

Unleashing the Potent Antitumor Force: A Reactive Oxygen Species (ROS) Storm Formation by Ultrasound-Activatable Metal Nanosonosensitizers.

Reactive oxygen species (ROS)-mediated tumor therapy, which induces oxidative stress damage for precise oncolysis, represents a novel antitumor strategy. However, the overexpression of glutathione (GSH) in the tumor microenvironment (TME) forms a strong antioxidant barrier, rapidly scavenging ROS and repairing oxidative damage, thus limiting the efficacy of conventional ROS-based therapies. Metal nanosonosensitizers, activated by ultrasound and penetrating deeply into the TME, offer a promising solution when combined with glutaminase1 (GLS1) inhibitors to overcome GSH-mediated defenses. This s

Sono-tribocatalytic therapy provokes PANoptosis in cancer cells.

The rapid progress of interdisciplinary medicine has significantly boosted ultrasound-based therapeutic modalities and strategies. The sonosensitizers, as core components of traditional sonodynamic therapy, have drawn escalating research focus for their unique therapeutic performance, but their clinical translation remains severely hampered by critical bottlenecks of biodegradability, required semiconductor characteristic, and long-term biosafety risks, which underscore an urgent demand for innovative sonosensitizers coupled with breakthrough therapeutic modalities to revolutionize ultrasound-

Ultrasound-triggered dual-mode therapy for heart failure via synchronized electrical stimulation and nitric oxide release.

The multifaceted pathophysiology of heart failure demands therapeutic strategies that can concurrently address its electrophysiological and metabolic components. Here, we report a lead-free and battery-free piezoelectric nanoplatform that, under ultrasound irradiation, serves as a dual-mode platform to simultaneously generate localized electrical stimulation and trigger spatiotemporally controlled nitric oxide (NO) release. In a mouse model of pressure-overload-induced heart failure, this ultrasound-driven regimen restored cardiac contractile function, reversed pathological remodeling, and sup

Hyaluronic acid-decorated MOF nanoplatform loaded with berbamine elicits immunogenic ferroptosis through a sonodynamic/chemodynamic/chemotherapy synergy in triple-negative breast cancer.

Chemoresistance, inadequate tumor targeting, and severe systemic toxicity remain key barriers to effective breast cancer treatment. Ferroptosis-inducing strategies have emerged as a promising therapeutic avenue, yet their efficacy is frequently compromised by the intrinsic antioxidant defense system and insufficient immune activation within the tumor microenvironment (TME). Herein, a hyaluronic acid (HA)-functionalized, berbamine (BBM)-loaded iron-based metal-organic framework (MOF) nanoplatform (HA-FeTCPP@BBM, HAFTB) was engineered. This nanosystem executes a "three-in-one" synergistic strate

Engineering nanozyme immunomodulator with magnetic targeting effect for cascade-enzyodynamic and ultrasound-reinforced metallo-immunotherapy in prostate carcinoma.

Conventional immunotherapy exhibits low response rates due to the immunosuppressive tumor microenvironment (TME). To overcome this limitation, this study introduces ZFPG nanoparticles (ZFPG NPs) with ZnFe2O4@Pt cores and glucose oxidase (GOx) shells. The ZFPG NPs possess five-enzyme activities, good sonosensitivity, and remarkable magnetic targeting properties, which facilitate sono-metallo-immunotherapy for prostate cancer treatment in male mice. Specifically, the magnetic targeting ability effectively improves their accumulation in tumors while still showing enrichment in the liver and kidne

Ultrasound-Driven Coassembly of Anticancer Drugs into Carrier-Free Particles.

The evolution of drug resistance in tumor malignancies has necessitated advancements in anticancer drug therapy. Drug combination therapy, which can burden cancer progression at multiple target sites, has been used to address drug resistance and includes the coencapsulation of synergistic drugs within nanoparticle carriers. However, the use of organic and inorganic carriers can lead to additional material-induced safety concerns, including inflammation and antibody formation. Herein, we report an ultrasound-driven approach to combine synergistic anticancer drugs into carrier-free particles. Ve

Abdominal ultrasound stimulation alleviates DSS-induced colitis and behavioral disorders in mice by mediating the microbiota-gut-brain axis balance.

Inflammatory bowel disease (IBD) has the potential to induce neuroinflammation, which may increase the risk of developing neurodegenerative disorders. Ultrasound stimulation to the abdomen is a potential treatment for dextran sulfate sodium (DSS)-induced acute colitis. The present study aimed to investigate whether abdominal low-intensity pulsed ultrasound (LIPUS) can alleviate DSS-induced neuroinflammation through the microbiota-gut-brain axis. Male mice were fed DSS to induce ulcerative colitis. LIPUS stimulation was then applied to the abdomen at intensities of 0.5 and 1.0 W/cm2. Mouse bio

Delivering Focused Ultrasound to Intervertebral Discs Using Time-Reversal.

Chronic low back pain causes more disability worldwide than any other condition and is thought to arise in part through loss of biomechanical function of degenerate intervertebral discs (IVDs). Current treatments can involve replacing part or all of the degenerate IVDs by invasive surgery. Our vision is to develop a minimally invasive approach in which high intensity focused ultrasound (HIFU) is used to mechanically fractionate degenerate tissue in an IVD; a fine needle is then used to first remove the fractionated tissue and then inject a biomaterial able to restore normal physiologic functio

Development of Low Frequency (20-100 kHz) Clinically Viable Ultrasound Applicator for Chronic Wound Treatment.

This paper details the systematic approach used to develop a viable clinical prototype of a therapeutic ultrasound applicator and discusses the rationale and deliberations that led to the design strategy. The applicator was specifically devised to treat chronic wounds and-to the best of the author's knowledge-is the first truly wearable device with a proven record of reducing healing time, directly translating to a reduction of healthcare costs. The prototype operates in the kHz (20-100) range of frequencies and uses noncavitational and nonthermal levels of ultrasound energy. Hence, in the abs
