Development and prospect of treating Alzheimer's disease using the glymphatic system.

Alzheimer's disease (AD) is a common neurodegenerative disorder with progressive cognitive decline and typical amyloid-β (Aβ) and tau pathologies, whose pathogenesis remains unclear. The glymphatic system, a core cerebral waste clearance pathway, has become a critical target for AD pathogenic research and novel therapy development. This review explores the glymphatic system's structure and function, illustrates its correlation with AD pathogenesis, summarizes targeted therapies, and highlights research limitations and future directions to facilitate AD translational research. Nutritional modulation of the glymphatic system: mechanistic insights and clinical implications.

The glymphatic system is a brain-wide perivascular clearance pathway mediated by aquaporin-4 (AQP4) water channels at astrocytic endfeet and plays a key role in eliminating neurotoxic proteins, including amyloid-β, tau, and α-synuclein. Impaired glymphatic function has been implicated in neurodegenerative disorders such as Alzheimer's and Parkinson's diseases. While sleep and physical activity are established modulators of glymphatic activity, the role of nutrition remains less clearly defined. This review summarizes current evidence on how nutritional factors may influence glymphatic-relevant

Correlation Between Dietary Nutrition and Glymphatic System Activity in Healthy Participants.

Background Dietary nutrition is an important approach to the prevention and treatment of the physical and mental states of humans. Nowadays, many studies show the neuroprotective and antioxidative effects of nutrients, such as B vitamins, zinc, and iron, on the central nervous system (CNS). However, there were no studies focusing on the relationships between the serum concentration of nutrients and the brain glymphatic system activity and macroscopic waste clearance system, including reactive oxygen species. Objectives This study tries to evaluate the relationships between them using diffusion

Interaction Between the Glymphatic System and α-Synuclein in Parkinson's Disease.

The glymphatic system contributes to the clearance of amyloid-β from the brain and is disrupted in Alzheimer's disease. However, whether the system is involved in the removal of α-synuclein (α-syn) and whether it is suppressed in Parkinson's disease (PD) remain largely unknown. In mice receiving the intranigral injection of recombinant human α-syn, we found that the glymphatic suppression via aquaporin-4 (AQP4) gene deletion or acetazolamide treatment reduced the clearance of injected α-syn from the brain. In mice overexpressing the human A53T-α-syn, we revealed that AQP4 deficiency accelerate

Glymphatic System Impairment in Alzheimer's Disease and Idiopathic Normal Pressure Hydrocephalus.

Approximately 10% of dementia patients have idiopathic normal pressure hydrocephalus (iNPH), an expansion of the cerebrospinal fluid (CSF)-filled brain ventricles. iNPH and Alzheimer's disease (AD) both exhibit sleep disturbances, build-up of brain metabolic wastes and amyloid-β (Aβ) plaques, perivascular reactive astrogliosis, and mislocalization of astrocyte aquaporin-4 (AQP4). The glia-lymphatic (glymphatic) system facilitates brain fluid clearance and waste removal during sleep via glia-supported perivascular channels. Human studies have implicated impaired glymphatic function in both AD a

Glymphatic system activity in functional movement disorder - a link to widespread pain.

AIM OF THE STUDY: The study aimed to evaluate glymphatic function in functional movement disorder (FMD) and examine its clinical correlates. CLINICAL RATIONALE FOR THE STUDY: The glymphatic system plays a key role in brain homeostasis by facilitating perivascular clearance of interstitial fluid and metabolic waste, particularly during sleep. The diffusion tensor imaging analysis along the perivascular space (DTI-ALPS) index provides a non-invasive magnetic resonance imaging-derived (MRI-derived) proxy of glymphatic system activity. The involvement of the glymphatic system in FMD is unknown.

The interplay between chronobiology, neuroinflammation, and the glymphatic system in neurodegenerative diseases.

Neurodegenerative diseases are traditionally characterized by the progressive accumulation of misfolded proteins and chronic neuroinflammation. However, emerging evidence indicates that the pathogenesis of these disorders is increasingly recognized to involve the disruption of temporal homeostasis. This conceptual review examines the highly interconnected, bidirectional crosstalk between the molecular circadian clock, glial-mediated neuroimmunity, and the macroscopic glymphatic clearance system. Under physiological conditions, the intrinsic circadian machinery helps maintain microglia in a qui

Novel Therapeutic Insights Into Alzheimer's Disease: Glymphatic System and Meningeal Lymphatic Vessels.

With the acceleration of global aging, the relationship between Alzheimer's disease (AD) and the glymphatic system (GS) has become a research hotspot in the field of neuroscience in recent years. Traditionally, the central nervous system was thought to lack a lymphatic system; however, research over the past decade has overturned this view. Studies have revealed the existence of GS and meningeal lymphatic vessels (mLVs) in the brain, which clear metabolic waste (such as amyloid-β and tau proteins) through the exchange of cerebrospinal fluid (CSF) and interstitial fluid (ISF). Dysfunction of th

Effect of organismal rhythmic activity on Aβ clearance by the glymphatic system.

Abnormal deposition of β-amyloid (Aβ) is a significant pathological feature of neurodegenerative diseases, particularly Alzheimer's disease (AD). The glymphatic system (GS) plays a crucial role in Aβ clearance. Various rhythmic activities of the organism dynamically influence Aβ clearance by modulating GS function. In this paper, we systematically review the mechanisms linking cardiovascular rhythms, respiratory rhythms, neural rhythms, circadian rhythms, and exercise patterns to Aβ clearance via the GS. Cardiovascular rhythms affect cerebral perfusion pressure and vascular pulsation to regula

Association of glymphatic system disturbance with neural dysfunction in amyotrophic lateral sclerosis.

BACKGROUND: Formation and aggregation of pathological proteins in the brain constitutes a critical hallmark of amyotrophic lateral sclerosis (ALS). However, the role of the glymphatic system in the clearance of pathological proteins in ALS remains unclear. The purpose of this cross-sectional study was to evaluate glymphatic system disturbance in ALS and its relation to neural function. METHODS: This study included 38 healthy controls (HCs) and 30 patients with ALS who underwent diffusion tensor imaging (DTI) and resting-state functional magnetic resonance imaging (rs-fMRI).

Glymphatic system impairment in multiple sclerosis: relation with brain damage and disability.

Recent evidence has shown the existence of a CNS 'waste clearance' system, defined as the glymphatic system. Glymphatic abnormalities have been described in several neurodegenerative conditions, including Alzheimer's and Parkinson's disease. Glymphatic function has not been thoroughly explored in multiple sclerosis, where neurodegenerative processes are intermingled with inflammatory processes. We aimed to investigate glymphatic system function in multiple sclerosis and to evaluate its association with clinical disability, disease course, demyelination and neurodegeneration, quantified using d

Glymphatic System as a Gateway to Connect Neurodegeneration From Periphery to CNS.

The classic concept of the absence of lymphatic vessels in the central nervous system (CNS), suggesting the immune privilege of the brain in spite of its high metabolic rate, was predominant until recent times. On the other hand, this idea left questioned how cerebral interstitial fluid is cleared of waste products. It was generally thought that clearance depends on cerebrospinal fluid (CSF). Not long ago, an anatomically and functionally discrete paravascular space was revised to provide a pathway for the clearance of molecules drained within the interstitial space. According to this model, C
