A Deep Clinical and Biochemical Characterization of a Patient With Combined Malonic and Methylmalonic Aciduria (CMAMMA).

Combined malonic and methylmalonic aciduria (CMAMMA) is an inborn error of metabolism caused by a deficiency in mitochondrial malonyl-CoA synthetase, the enzyme responsible for activating malonic acid (MA) to malonyl-CoA, a precursor of lipoic acid. The clinical phenotype is highly heterogeneous, ranging from asymptomatic cases to severe neurological impairment. We describe a patient affected by CMAMMA. The patient presented in the neonatal period with hyperinsulinemic hypoglycemia. Urinary organic acid analysis revealed elevated levels of both malonic and methylmalonic acids. The diagnosis of

In situ Malone antegrade continence enema in 127 patients: a 6-year experience.

PURPOSE: The initial description of the Malone antegrade continence enema (MACE) relied on a reversed, tunneled and reimplanted appendix. In 1999 we reported our in situ technique that uses windows developed in the appendiceal mesentery for imbrication. We present our long-term results. MATERIALS AND METHODS: From 1997 to 2003, 168 patients were identified who had undergone a MACE procedure. An in situ technique was performed in 76 females and 51 males. Average patient age at the time of surgery was 9.6 years (range 2.9 to 28.4). Diagnoses included myelomeningocele in 116 cases, lipomeningoce

[Analysis of the pollutant in purified diethyl malonate by on-line LG-GC/FTIR method].

In some cases, the use of conventional GC/FTIR technique is limited due to its lower detectability to micro-components in organic mixtues in comparison to GC/MS. In this paper, an integration of LG/GC using retention gap technique with partially concurrent solvent evaporation, as a efficient chromatographic system for sample preconcentration and preseparation, was coupled into FTIR system in order to overcome the detectability problems of GC/FTIR analysis. The applicability of the on-line LG-GC/FTIR method is demonstrated by analysis of the isomers of divinylbenzene in purified diethyl malonat

Solvent extraction of scandium from malonic acid with high molecular-weight amines.

Scandium is quantitatively extracted with 4% Amberlite LA-1 or Amberlite LA-2 in xylene at pH 2.5-5.5 from 0.1M malonic acid. Scandium is stripped from the organic phase with 0.5M hydrochloric acid and determined spectrophotometrically at 525 nm, as its complex with Alizarin Red S. Primene JM-T, tri-iso-octylamine, tributylamine and tribenzylamine have also been studied as extractants, but found to be unsatisfactory for various reasons. Xylene, toluene, benzene, chloroform, carbon tetrachloride, hexane, cyclohexane and kerosene have been studied as diluents. Xylene is found to be the most effi

Targeted LC-MS/MS Analysis of Malonic Acid Biomarkers in Clinical Laboratory Applications.

LC-MS/MS enables sensitive quantification of malonic acid (MA), methylmalonic acid (MMA), and ethylmalonic acid (EMA) across clinical matrices. MA provides emerging diagnostic value in metabolic assessments, while MMA remains the primary marker of functional vitamin B12 deficiency. EMA is essential for evaluating mitochondrial and fatty-acid oxidation disorders. Advances in extraction and chromatographic strategies continue to improve analytical performance for these biomarkers.

Local arterial administration of acidified malonate as an adjunct therapy to mechanical thrombectomy in ischemic stroke.

AIMS: Ischemic stroke is increasingly treated by mechanical thrombectomy (MT) with the more rapid and complete reperfusion of the ischemic tissue, enhancing patient outcome, compared to recombinant tissue plasminogen activator (rtPA) alone. Even so, there is still extensive brain infarction and disability following MT, which is exacerbated by ischemia-reperfusion injury (IRI) and other pathological processes during reperfusion. Hence, an adjunct therapy to MT that decreases IRI should enhance patient outcomes. METHODS AND RESULTS: To test this possibility, we adapted the transient middle cere

Usefulness of the Monti-Malone procedure as a reconstruction of the antegrade continence enema procedure: a case report.

BACKGROUND: The antegrade continence enema (ACE) procedure is effective for severe constipation in patients with spina bifida and can improve quality of life (QOL). The Monti-Malone procedure (MM), which is a method of creating an enema tract from the colon, has been reported as an alternative to the ACE procedure when the appendix cannot be used. We report the usefulness of MM as a reconstruction of the antegrade continence enema procedure. CASE PRESENTATION: Our patient was a 22-year-old man with congenital spina bifida and hydrocephalus. Ventriculoperitoneal (VP) shunt surgery was performe

Synthesis, Biological Activity and Action Mechanism Study of Novel Chalcone Derivatives Containing Malonate.

A series of novel chalcone malonate derivatives were synthesized and their antibacterial and antiviral activities were evaluated. All target compounds were characterized by spectral data. The results of antimicrobial bioassay showed that one compound (diethyl [3-(naphthalen-2-yl)-1-(3-nitrophenyl)-3-oxopropyl]propanedioate) showed excellent antibacterial activity against Xanthomonas oryzae pv. oryzae (Xoo), with an EC50 value of 10.2 μg/mL, which is significantly superior to bismerthiazol (71.7 μg/mL) and thiodiazole copper (97.8 μg/mL). At the same time, the mechanism of two compounds was con

Selective Inhibition of Succinate Dehydrogenase in Reperfused Myocardium with Intracoronary Malonate Reduces Infarct Size.

Inhibition of succinate dehydrogenase (SDH) with malonate during reperfusion reduces infarct size in isolated mice hearts submitted to global ischemia. However, malonate has toxic effects that preclude its systemic administration in animals. Here we investigated the effect of intracoronary malonate on infarct size in pigs submitted to transient coronary occlusion. Under baseline conditions, 50 mmol/L of intracoronary disodium malonate, but not lower concentrations, transiently reduced systolic segment shortening in the region perfused by the left anterior descending coronary artery (LAD) in op

Pre-operative risk factors for stomal stenosis with Malone antegrade continence enema procedures.

INTRODUCTION: The Malone antegrade continence enema (MACE) procedure remains an effective tool in providing independence to patients with refractory constipation. Appendiceal, ileal and colonic segments are all used depending upon the patient's individual anatomy and surgeon's preference. Unfortunately, MACE stomal stenosis (SS) remains a frustrating complication. It was hypothesized that SS is associated with the type of efferent limb used for MACE construction. OBJECTIVE: To perform a retrospective risk analysis of the pre-operative factors that correlated with postoperative stomal stenosis

Assessment of pediatric Malone antegrade continence enema (MACE) complications: Effects of variations in technique.

OBJECTIVE: The Malone antegrade continence enema (MACE) procedure is performed for patients with fecal incontinence and constipation who do not respond to medical therapy. The MACE procedure provides antegrade catheter access to the right colon and allows administration of enemas to help evacuate the bowel regularly. The objective of this study was to identify risk factors for surgical complications in children who undergo the MACE procedure. METHOD: Records from 97 pediatric patients who underwent MACE procedure were reviewed retrospectively. Data collected included age, sex, weight (obese/n

Use of Malone antegrade continence enema in patients with perineal colostomy after rectal resection.

PURPOSE: Abdominoperineal resection, with iliac colostomy, remains the gold standard treatment for very low-lying rectal cancer, but it alters patients' quality of life. Alternatives to iliac colostomy need to be experimented. Antegrade enemas via a cecal access (Malone operation) obtains a colonic emptying and improves continence for incontinent patients. Continence and quality of life after abdominoperineal resection and perineal colostomy associated to a Malone antegrade continence enema were studied. METHODS: After abdominoperineal resection for cancer, 18 patients had a digestive reconst

Modified cecal flap neoappendix for the Malone antegrade continence enema procedure: a novel technique.

PURPOSE: We report our experience with the creation of a tubularized cecal flap to construct a neoappendix. This flap effectively serves as an alternative means of creating a conduit for antegrade continence enemas in patients who have either an absent or an unusable appendix. MATERIALS AND METHODS: We reviewed our results from our initial 4 patients in whom we used this technique. A neoappendix is created from a medially based flap of distal cecum that is tubularized over a catheter and then buried in a seromuscular tunnel to create an antireflux mechanism. A skin stoma is then created. RES

Photo-induced cytotoxicity of malonic acid [C(60)]fullerene derivatives and its mechanism.

The biological activities of fullerenes have attracted extensive attention in recent years. The aim of this paper is to study the relation of the photo-induced cytotoxicity of fullerene derivatives to their chemical structures as well as the possible cellular mechanism involved in the photocytotoxicity. Three C(60) derivatives with two to four malonic acid groups (DMA C(60), TMA C(60) and QMA C(60)) were prepared and the cytotoxicity of these compounds against HeLa cells was determined by MTT. Cell cycle was measured by flow cytometry. The results showed that the cytotoxicity of the malonic ac
