Standardization of tissue handling in veterinary pathology: How fixation affects morphological and molecular examination results.

Variables in the fixation and processing of histological specimens and storage of formalin-fixed paraffin-embedded (FFPE) tissues affect the outcomes of morphological and molecular examinations. The present study investigated the influence of unbuffered formalin fixation and prolonged fixation on histomorphology, immunohistochemistry, polymerase chain reaction (PCR) analysis, and DNA quality in FFPE tissues, which are essential issues in veterinary diagnostic pathology. Fixation of tissues with 10% unbuffered formalin resulted in increased intercellular spaces and decreased color contrast in h

Talar Arthroscopic Reduction and Internal Fixation (TARIF): A Novel All-Inside Soft-Tissue-Preserving Technique.

BACKGROUND: Talar arthroscopic reduction and internal fixation (TARIF) is an alternative approach for the operative fixation of talar fractures that may be utilized instead of more traditional open approaches such as medial, lateral, or even dual anterolateral. The TARIF approach allows for nearly anatomic fracture reduction and fixation of talar neck, body, and posterior dome fractures while minimizing the soft-tissue stripping and vascular injury associated with the standard anterolateral approach. DESCRIPTION: Following initial closed fracture reduction and any associated procedures, we re

Assessing Tissue Fixation Time and Quality with Label-free Mid Infrared Spectroscopy and Machine Learning.

Objectives: This work investigates whether changes in a biospecimen's molecular composition from formaldehyde fixation drive changes in the mid infrared (MID-IR) spectrum. Our ultimate goal was to develop an analytical metrology that could be used to accurately determine the fixation time of a tissue sample as a surrogate to overall tissue quality. Methods: Multiple unstained formalin-fixed paraffin-embedded tissue samples were scanned with an MID-IR microscope to identify a molecular fingerprint of formaldehyde fixation. The fixation specific patterns were then mined to develop a predictive m

Effect of formalin fixation on measured concentrations of deposited gadolinium in human tissue: an autopsy study.

BACKGROUND: Generally, studies of gadolinium (Gd) deposition in humans measure concentration by analyzing formalin fixed postmortem tissue. However, the effect of formalin fixation on measured Gd concentration has not been well investigated. PURPOSE: To evaluate the effect of fixation by comparing Gd concentration in fresh versus formalin-fixed postmortem human tissues. MATERIAL AND METHODS: Fresh samples of bone and skin were collected from autopsy cases with previous exposure to Gd-based contrast agents (GBCAs). The type of GBCA administered, dose, and estimated glomerular filtration rate

Effects of Formalin Fixation on Trace Element Concentrations in Bottlenose Dolphin (Tursiops truncatus) Tissues.

Odontocetes are considered ideal sentinel species to monitor environmental trace element concentrations. Although frozen tissues are preferable for trace element analysis, formalin-fixed tissues are often the only samples available; however, it is uncertain whether formalin fixation alters tissue trace element concentrations. To explore whether formalin-fixed tissues could be utilized for toxicology studies, concentrations of 14 trace elements (arsenic [As], cadmium, cobalt, chromium, copper, iron, mercury, manganese, nickel, lead, selenium, tin, vanadium, and zinc [Zn]) were measured in froze

Systematic evaluation of PAXgene® tissue fixation for the histopathological and molecular study of lung cancer.

Whilst adequate for most existing pathological tests, formalin is generally considered a poor DNA preservative and use of alternative fixatives may prove advantageous for molecular testing of tumour material; an increasingly common approach to identify targetable driver mutations in lung cancer patients. We collected paired PAXgene® tissue-fixed and formalin-fixed samples of block-sized tumour and lung parenchyma, Temno-needle core tumour biopsies and fine needle tumour aspirates (FNAs) from non-small cell lung cancer resection specimens. Traditionally processed formalin fixed paraffin wax emb

Effect of formalin fixation on pcr amplification of DNA isolated from healthy autopsy tissues.

The aim of this study is to investigate the effects of formalin fixation on the degradation of DNA molecules in five different healthy tissues exempted during the autopsy, as well as the selection of the method that is most suitable for the DNA isolation. Heart muscle, liver, brain, lung and kidney tissue obtained from the healthy people who suddenly died from a violent death were used. The parts of tissue were fixed in 10% phosphate-buffered formalin as well as in 4% unbuffered formalin at room temperature. Morphology of tissue was studied using H&E staining. The DNA was isolated 6 h, 1-7 day

Formalin Fixation and Cryosectioning Cause Only Minimal Changes in Shape or Size of Ocular Tissues.

Advances in imaging have made it increasingly common to study soft tissues without first embedding them in plastic or paraffin and without using labels or stains. The process, however, usually still involves fixation and cryosectioning, which could deform the tissues. Our goal was to quantify the morphological changes of ocular tissues caused by formalin fixation and cryosectioning. From each of 6 porcine eyes, 4 regions were obtained: cornea, equatorial and posterior sclera, and posterior pole containing the optic nerve head. Samples were imaged using visible light microscopy fresh, 1-minute

Detection of viral RNA from paraffin-embedded tissues after prolonged formalin fixation.

BACKGROUND: Isolating amplifiable RNA from formalin-fixed, paraffin-embedded (FFPE) tissues is more difficult than isolating DNA because of RNases, chemical modification of the RNA, and cross-linking of nucleic acids and proteins. Tissues containing infectious disease agents that require biosafety level (BSL)-3 and -4 necessitate fixation times of 21 and 30 days, respectively. OBJECTIVE: To improve procedures for extracting RNA from these FFPE tissues and detect the RNA with the more sensitive TaqManbased reverse transcriptase (RT)-PCR. STUDY DESIGN: Through a single modification of a commer

Long-term fixation impact on archived human nervous tissues for sequencing-based transcriptomics.

The advent of transcriptomics technologies compatible with formalin-fixed paraffin-embedded tissues has enabled greater opportunities for scientists to interrogate the transcriptome of archived tissues, especially those from rare neurodegenerative diseases. Tissues from brain banks are often placed in fixative for weeks or years due to limitations in resources and storage space. We investigated the impact of long-term fixation for sequencing-based transcriptomics in human formalin-fixed paraffin-embedded lumbar spinal cord and hypothalamic tissues sourced from the Sydney Brain Bank.
