BACKGROUND: Sand flies are vectors of various pathogens, primarily Leishmania and phleboviruses. The effectiveness of disease control strategies can be assessed using serological tests that measures antibodies against sand fly saliva as a marker of host exposure and a proxy biomarker of sand fly-borne pathogens transmission. Recently, we proposed novel recombinant salivary proteins to evaluate exposure of dogs to Phlebotomus tobbi and P. papatasi. The aim of this study was to test vector and host specificity of these recombinant proteins.
METHODS: Recombinant salivary proteins of P. papatasi

The Membrane-Associated Proteins in Eicosanoid and Glutathione metabolism (MAPEG) superfamily comprises integral membrane proteins involved in lipid metabolism, detoxification, and redox regulation. While extensively characterised in eukaryotes, bacterial MAPEG homologs remain poorly understood. In this study, we conducted an in silico analysis of bacterial MAPEG proteins and performed a comparative investigation with selected eukaryotic homologs to assess structural conservation and evolutionary relationships. Sequence alignment and phylogenetic reconstruction showed that the bacterial protei

Gamete recognition proteins are expressed on the surfaces of sperm and eggs, where they mediate interactions between gametes. The genetic basis for gamete recognition proteins, as well as their structure and interactions, have yet to be fully resolved. Using a new high-quality de novo genome assembly for the sea urchin Mesocentrotus franciscanus, we investigated the genomic structure, expression, and protein forms of several gamete recognition proteins: sperm bindin, egg receptor for sperm (HSP110), and egg bindin receptor (EBR1), as well as the receptor for egg jelly (REJ) and its paralogs. Mitochondrial matrix Ca2+ concentration ([Ca2+]m) is theorized to be an essential regulator of mitochondrial metabolism by positively regulating key mitochondrial dehydrogenases. However, ablation or functional inhibition of the mitochondrial calcium uniporter channel (mtCU) fails to significantly perturb basal metabolism and is largely phenotypically silent in the absence of stress. Here we demonstrate that MICU proteins, the reported gatekeepers of mtCU, function in coordination to impart calcium-dependent regulation to FADH2-dependent mitochondrial dehydrogenases through metabolon formation

AIMS: We investigated the role of Lgi3-4 proteins in cardiac electrophysiology, with a specific focus on IKur, and their potential contribution to the pathophysiology of atrial fibrillation (AF).
METHODS AND RESULTS: In human atrial tissue and heterologous cells, Lgi3 and Lgi4 interacted with KV1.5 channels. In a mouse model of AAV9-mediated cardiac-specific Lgi4 expression, sinoatrial and atrioventricular conduction were impaired, resulting in a prolonged QRS interval. Action potential repolarisation was delayed in atrial and ventricular cardiomyocytes from Lgi4 mice. In HEK293 cells, Lgi3-4

The development of functional foods enriched with nutraceuticals offers a targeted strategy to enhance gut health and overall well-being. Protein-based matrices are increasingly explored for nutraceutical applications due to their biocompatibility, biodegradability, and ability to interact with diverse bioactive compounds. However, the rational selection of compatible protein-bioactive combinations, crucial to ensure nutraceutical efficacy, is constrained by inadequate structure-based screening. This study investigated the potential of upcycled jojoba proteins (JP) as sustainable functional ma

The adaptability and persistence of Staphylococcus aureus in diverse host environments may be influenced by its genetic repertoire, including RNA-binding proteins (RBPs). These proteins play critical roles in regulating post-transcriptional processes; however, their significance in bacteria, particularly in pathogenic strains, remains poorly understood. This gap necessitates a comprehensive exploration of RBPs in bacterial pathogens such as S. aureus better to understand their potential contributions to virulence and adaptability. In this study, a bioinformatics-based analysis of the genomes a

Modern military performance increasingly depends on cognitive resilience and neurological health, making nutrition a strategic determinant of combat readiness. This review integrates nutritional neuroscience and military medicine to explore duckweed as a next-generation neuro-nutritional resource. Duckweed species possess protein content (25-45% dry weight) and a complete essential almost amino acid (EAA) profile that may contribute to pathways involved in neurotransmitter synthesis, neuroplasticity, and stress adaptation. Beyond amino acids, duckweed delivers neuroprotective compounds such as

Thermal processing of egg white proteins (EWPs) often results in a fishy odor in food products. Here, ternary composite systems (ECX) were developed using caseins (CNs) and xylitol (XY), aided by high-intensity ultrasound (HIU), to mitigate off-flavor formation. Volatile organic compounds (VOCs) were profiled by GC-IMS, while sensory changes were evaluated using an E-nose and E-tongue coupled with multivariate analysis. The results showed that 46 volatiles were detected, including 18 odor-active compounds. Notably, applying HIU to ECX for 10 min after heating significantly reduced trimethylami

Heat shock proteins (HSPs) are a family of conserved molecular chaperons present in both prokaryotic and eukaryotic species, playing a crucial role in maintaining cellular proteostasis and enhancing stress resilience. HSPs have a multitude of roles in regulating cell signaling transduction, antioxidant defenses, apoptosis, and protein folding, thereby contributing to overall cellular homeostasis. Insulin resistance is characterized by elevated oxidative stress, dysregulated pro-inflammatory signaling, and impaired cellular stress response, ultimately leading to deficient glucose uptake in skel

Originally identified as a chromatin domain insulator binding protein, Drosophila melanogaster BEAF (Boundary Element-Associated Factor of 32 kDa) is predominantly associated with transcription start sites (TSSs) of housekeeping genes. It was previously shown that BEAF directly interacts with the transcription factor Serendipity-δ (Sry-δ). TSS-proximal BEAF can interact with distantly bound Sry-δ to activate a reporter gene. However, it is unknown if the ability of BEAF to communicate with transcription-associated factors from a distance is a common property. Here, we use yeast 2-hybrid assays

Esca is a major grapevine trunk disease causing devastating economic losses in vineyards. This complex disease results from trunk colonization by fungi such as Phaeomoniella chlamydospora (P.ch). Because the characterization of early molecular events occurring during infection is difficult to perform directly in grapevine trunks, we previously developed a procedure based on stomatal movement to investigate early signaling events during pathogen perception. Assays performed on epidermal peels revealed that cell wall-associated proteins, but not polysaccharides, from P.ch induced stomatal closur

O-glycosylation of Notch epidermal growth factor-like (EGF) repeats has long been studied in the context of protein folding, secretion from cells, and protein function. Originally, the protein O-glucosyltransferase, POGLUT1, was thought to be the only enzyme adding an O-glucose modification to EGFs, specifically to a serine between cysteine one and two of a six cysteine containing EGF repeat. The POGLUT1 O-glucose can be elongated on Notch EGFs with xyloses by GXYLT1/2 and XXYLT1, forming a trisaccharide. Mouse knockouts of Poglut1 are embryonic lethal with Notch1-related phenotypes. Recently,

We report a site-specific PEGylation strategy to stabilize insulin-like growth factor 2 (IGF2) as a functional ligand for insulin-like growth factor 2 receptor (IGF2R)-based lysosome-targeting chimera (LYTAC) systems. This chemical modification improves ligand folding and solubility, enabling efficient LYTAC-mediated epidermal growth factor receptor (EGFR) degradation across multiple cell lines and significant tumor suppression (∼70%) in vivo with favorable safety. This study provides a general chemical approach for ligand optimization in lysosome-targeted protein degradation, providing new id

As a trace element, copper plays a vital role in regulating cellular homeostasis by serving as a cofactor for various enzymes. Therefore, maintaining an adequate copper concentration inside the cell is crucial. Even moderate dysregulation of homeostasis can lead to cytotoxicity, protein aggregation, resulting in cell death. One of the significant cellular consequences in Saccharomyces cerevisiae is the inhibition of Sec61-mediated protein translocation in the secretory pathway, resulting in the accumulation of the cytosolic form of precursor secretory proteins, such as Gas1 and CPY, in the cyt

Maternal consumption of alcohol and drugs during pregnancy can compromise neural development with long-lasting impact on individuals' health. The inhibitor of growth (ING) family of proteins is an epigenetic regulator that plays a central role in fetal brain development, contributing to neural stem cell maintenance, neuronal differentiation, and the regulation of genes involved in brain morphogenesis. Given the susceptibility of the developing nervous system to epigenetic dysregulation induced by alcohol and drugs, this narrative study aims to summarize literature evidence with the hypothesis

Fungal mycelial proteins are promising sustainable protein sources, yet their nutritional utilization is often limited by structural constraints. This study systematically evaluated the effects of protein isolation on the proteomic composition, gastrointestinal digestion behavior, amino acid utilization, and bioactivity of Pleurotus citrinopileatus mycelial proteins. Quantitative proteomics identified 3591 proteins, of which 3374 were shared between mycelial flour (PCMF) and protein isolate (PCMPI), indicating that PCMPI primarily represents the soluble proteome fraction. The gut, brain, fat body, and various glands, although rich in "CSPs", are not involved in chemosensory functions. We propose renaming the "CSPs" to "4CSPs" (4-Cysteine Soluble Proteins) to clarify their role, avoiding confusion with proteins also found in wings, hemolymph, and eggs. This report highlights studies related to insecticide resistance and lipid transport and expands on potential intracellular functions, strongly indicating non-chemosensory features of 4CSPs. Additionally, we show a significant correlation between 4CSPs and mucins, translation initiation factors, and actin complex

Huntington's disease (HD) is a life-altering genetic neurodegenerative disorder, with cognitive, motor, and psycho-social effects that have consequential impacts on the individuals and their families. While current treatments improve disease symptoms, there are no FDA-approved therapies that prevent disease progression. Converging lines of evidence from human GWAS and mouse models point to DNA repair and handling (R/H) proteins as promising therapeutic targets due to their ability to modulate somatic expansion of the CAG repeat of HTT. The roles of DNA R/H HD modulator proteins are incompletel
