PURPOSE: Process Analytical Technology (PAT) tools are increasingly applied to improve monitoring of biopharmaceutical processes; however, few techniques have explored their use in liquid bioprocesses based on insect cell cultures. This work evaluated whether rheological parameters could be explored as monitoring indicators for biochemical variables during virus-like particle (VLP) production using the insect cell-baculovirus expression system.
METHODS: Batch bioreactor cultivations of Spodoptera frugiperda (Sf9) cells were conducted under non-infected conditions and during rabies and SARS-Co

BACKGROUND: Mucoactives such as hypertonic saline (HTS) and carbocisteine are widely used in the treatment of bronchiectasis, though there is insufficient evidence to support their use. The aim of this mechanistic sub-study, embedded within the CLEAR trial, was to characterise the properties of sputum from patients with bronchiectasis and to assess whether treatment with HTS and/or carbocisteine altered these properties.
METHODS: In CLEAR, patients were randomised to receive HTS, carbocisteine, HTS plus carbocisteine or standard care, and sputum samples were collected at randomisation (baseli

Controlling the rheological properties of liquids allows for the regulation of effective movement, transport of substances, and processes in biological systems. This work presents an experimental investigation into the influence of the proton-exchange polymer membrane Nafion on the surface tension coefficient (STC) of distilled water, aqueous solutions of two methylene blue (MB) forms, and ascorbic acid (AA). Immediately upon membrane immersion in the solutions, a sharp decrease in the surface tension of distilled water, as well as of the oxidized and reduced forms of MB, occurs. The observed

This study investigated the microstructural, rheological and tribological properties of model W/W emulsions composed of dextran (D) and poly(ethylene oxide) (P) at a fundamental level. Rheological analysis revealed that increasing the P concentration, [P], resulted in increased viscosity (η), whilst increasing the D concentration, [D], intensified shear-thinning behaviour, likely due to changes in the size of D-based droplets. Confocal laser scanning microscopy (CLSM) demonstrated a significant increase in the average droplet size with higher [D] or [P]. A striking tribological result was that

Gaucher Disease (GD) is a rare genetic disorder characterized by a deficiency in the enzyme glucocerebrosidase, leading to the accumulation of glucosylceramide in various cells, including red blood cells (RBCs). This accumulation results in altered biomechanical properties and rheological behavior of RBCs, which may play an important role in blood rheology and the development of bone infarcts, avascular necrosis (AVN) and other bone diseases associated with GD. In this study, dissipative particle dynamics (DPD) simulations are employed to investigate the biomechanics and rheology of blood and

Injectable hyaluronic acid (HA) hydrogels have been popularized in facial aesthetics as they provide a long-lasting effect, low risk of complications, allergenicity tests are not required before application and can be easily removed by the action of hyaluronidases. On the other hand, the development of these systems requires in-depth studies of chemical mechanisms involved in hydrogel formation. Ideal dermal fillers should temporarily fluidize during extrusion through the needle and quickly recover their original shape after application. Hydrogels with more elastic properties, for example, are

BACKGROUND: Pregnancy is a dynamic process associated with changes in vascular and rheological resistance. Maternal maladaptation to these changes is the leading cause of pregnancy complications such as preeclampsia.
OBJECTIVE: This study aimed to assess the hemorheological alterations in pregnancies with a high risk for preeclampsia in the first trimester.
METHODS: Ninety-two pregnant women were allocated into the high preeclampsia risk group (37 cases) and control groups (55 cases). Plasma and whole blood viscosity and red blood cell morphodynamic properties, including deformability and ag

In rotating Hele-Shaw flows, centrifugal force acts, and the interface separating two viscous fluids becomes unstable, driven by the density difference between them. Complex interfacial structures develop where fingers of various shapes and sizes grow, and compete. These patterns have been well studied over the last few decades, analytically, numerically, and experimentally. However, one feature of the pattern-forming dynamics of much current interest has been underappreciated: the role of surface rheological stresses in the deformation, and time evolution of the fluid-fluid interface. In this

Stimuli-responsive materials or smart materials are designed materials whose properties can be changed significantly by applying external stimuli, such as stress, electric or magnetic fields, light, temperature, and pH. We report the linear and nonlinear rheological properties of thermoresponsive composite gels based on submicron-sized hematite pseudocube-shaped particles and a triblock copolymer Pluronic F127 (PF127). These novel composites form hard gels at an elevated temperature of 37 °C. For certain concentrations (<20 w/v. %) of hematite pseudocubes in 17.5 w/v. % of PF127, the gel stren

In situ characterization of minute amounts of fluids that rapidly change their rheological properties is a challenge. In this paper, the rheological properties of fluids were evaluated by examining the behavior of magnetic nanorods in a rotating magnetic field. We proposed a theory describing the rotation of a magnetic nanorod in a fluid when its viscosity increases with time exponentially fast. To confirm the theory, we studied the time-dependent rheology of microdroplets of 2-hydroxyethyl-methacrylate (HEMA)/diethylene glycol dimethacylate (DEGDMA)-based hydrogel during photopolymerization s

BACKGROUND: Dysphagia can lead to aspiration of oral feeds, thus causing pneumonia. Dysphagia is diagnosed by assessing the ability to swallow barium test feeds (BTF) of different viscosities. Dysphagia diet foods (DDF) are thickened as recommended by the National Dysphagia Diet (NDD) guidelines. To our knowledge, there are no published data evaluating if the viscosity of BTF or commercial DDF meet NDD guidelines.
METHODS: A TA1000 rheometer (TA Instruments; New Castle, DE) measured dynamic viscosity of BTF and DDF using creep transformation under controlled stress. Thin DDF studied included

Rheological experiments have shown that canine submaxillary mucin (CSM) forms gels in aqueous solution at low ionic strength and in 6M GdnHCl. Examination of specimens of intact CSM and also its subunits prepared by reduction and carboxymethylation showed that the presence of lipid increases the gel-forming capability, probably as a result of enhancement of the intermolecular hydrophobic interactions. The rheological evidence for gelation is that substantially larger values of the oscillatory storage modulus, G' (omega), and the dynamic complex viscosity, eta*(omega), are observed for lipid-co

The noninvasive detection of deep venous thrombosis (DVT) in pregnancy is a significant problem. The Hemodynamics AV-1000 uses light reflection rheology in superficial capillaries to detect DVT but has not been evaluated adequately in pregnancy. Four hundred one pregnant women in the second and third trimesters were studied with the AV-1000; none had symptoms of DVT or thrombophlebitis, and all had uncomplicated pregnancy outcomes. AV-1000 reflection curves were related to maternal age, gestational age, weight gain, Quetelet index and device sensitivity settings. No statistically significant d

There is wide variation in the clinical manifestations of sickle cell disease (SCD) from one affected individual to another. Many investigators have sought to discern parameters that would explain this variability. In the present studies we have attempted to correlate the frequency of painful events and the extent of end organ failure in SCD with rheologic properties of packed suspensions of sickle cells, using a magneto-acoustic ball microrheometer developed in our laboratory. Using this device we have measured the steady-state viscosity, and the viscous and elastic moduli of cell suspensions

The flow of dense emulsions underlies applications from food and pharmaceutical processing to bioengineering, yet their rheology remains difficult to interpret under conventional volume-imposed conditions. Here, we change the control variable to osmotic-pressure using a recently developed instrument-the Capillarytron. This approach reveals a unified rheological structure where the osmotic pressure [Formula: see text], by controlling droplet deformation, sets a pressure-dependent jamming volume fraction. When expressed in terms of the distance to this jamming point, all rheological data-spannin

Food has been categorized as solid, molecular dispersion, colloidal (e.g., emulsions, gels, sols), and coarse dispersion that exhibit unique rheological behavior driven by polysaccharides, proteins, and lipids. These compounds exhibit various functional attributes such as texture, stability, and sensory quality. During processing, starch gelatinize, proteins denature and coagulate, and fat droplets interact with biopolymers to form viscoelastic or structured networks. These molecular and microstructural changes directly influence rheological behavior. Small-amplitude oscillatory shear (SAOS) r

Petroleum well drilling fluids are one of the most significant constituents in the subterranean drilling processes to meet an increasing global demand for oil and gas. Drilling fluids experience exceptional wellbore conditions, e.g. high temperature and high pressure that adversely affect the rheology of these fluids. Gas and oil well drilling operations have to adjourn due to changes in fluid rheology, since the drilling fluids may lose their effectiveness to suspend heavy particles and to carry drilled cuttings to the surface. The rheological properties of drilling fluids can be controlled b

This article describes molecular dynamics simulations of an ionic liquid (IL) confined between iron oxide surfaces under relatively high pressure and severe shearing, representative of a typical steel-steel lubricated contact. The simulations reveal the presence of hydrodynamic and thermal slip at the walls, despite the wetting nature of the fluid/wall interface. A crucial consequence of the temperature slip is the subsequent increase of the fluid temperature under shear, which modifies its effective rheology, resulting in saturation of the shear stress at high shear rates. Overall, this artic
