The hollowed root formation and 4'-deoxyflavone accumulation in Scutellaria baicalensis are associated with jasmonate (JAs) signaling, but the upstream regulators integrating these processes with drought tolerance remain unknown. This study investigated whether the 13-lipoxygenase gene SbLOX3 may act as a modulator. Transgenic plants overexpressing SbLOX3 or with RNAi suppression were exposed to polyethylene glycol-simulated and long-term soil drought stress. Metabolite profiling, gene expression analysis, root morphological measurements, and histological observations were conducted. Overexpre

The fermentation products were obtained through bidirectional solid fermentation technology, utilizing Scutellaria barbata D. Don as the medicinal substrate and Cordyceps kyushuensis as the fermentation strain. Five novel 4-methoxyglucosides, one new non-nucleoside cyclic peptide and 36 known compounds were isolated from the ethanol extract of the fermentation products. Notably, in the five novel glucosides, the 4-position hydroxyl group was modified to a methoxy group, likely due to enzymatic reactions during fermentation. In vitro assays revealed that compounds 4, 5, 16, 19, 20, 25, 27, 31,

AIMS: γ-aminobutyric acid (GABA) from reactive astrocytes is critical for the dysregulation of neuronal activity in various neuroinflammatory conditions. While Scutellaria baicalensis Georgi (S. baicalensis) is known for its efficacy in addressing neurological symptoms, its potential to reduce GABA synthesis in reactive astrocytes and the associated neuronal suppression remains unclear. This study focuses on the inhibitory action of monoamine oxidase B (MAO-B), the key enzyme for astrocytic GABA synthesis.
METHODS: Using a lipopolysaccharide (LPS)-induced neuroinflammation mouse model, we con

Inflammatory responses are key contributors to cancer cachexia and foster a complex cascade of biological outcomes. Baicalin is a natural compound derived from Scutellaria baicalensis that possesses anti-inflammatory properties in many diseases; therefore, the aim of this study was to verify whether baicalin could ameliorate cachexia in a CT26 adenocarcinoma-induced model. Tumour-bearing and control mice were injected with CT26 adenocarcinoma cells and phosphate-buffered saline (PBS), respectively, and baicalin was administered intraperitoneally for 15 days. During the study, food intake, body

Scutellaria barbata D. Don (SB) has long been used as a major component in numerous Chinese medical formulas to clinically treat various types of cancer. Previously, we reported that the extracts of SB were able to suppress colon cancer growth in vivo and in vitro, possibly by inducing cancer cell apoptosis and inhibiting cell proliferation and tumor angiogenesis. However, the anticancer mechanisms of its bioactive ingredients remain largely unclear. In the present study, using three human colon cancer cell lines SW620, HT-29 and HCT-8, the antitumor effect of different solvent fractions of SB

Zinc oxide nanoparticles (ZnONPs) are increasingly used in agriculture to stimulate plant growth and development, including under in vitro culture conditions. However, there is limited data on the effects of ZnONPs on the micropropagation of Scutellaria baicalensis Georgi. The pharmacological properties of this species make it a valuable medicinal plant. In Poland, it does not occur naturally but is cultivated for the production of herbal material. In vitro micropropagation is an effective method for obtaining genetically uniform plantlets. The aim of this study was to evaluate the effects of

We screened natural resources for estrogen receptor (ER)-activating and bone metabolism-promoting activities with the aim of finding potential treatments for osteoporosis. A screen of 1531 extracts from Ryukyu Arc plants using a luciferase reporter assay identified an 80% MeOH extract of Scutellaria rubropunctata var. rubropunctata (SRE) with dose-dependent ER transcription-promoting activity. Importantly, SRE had no proliferative effect on human breast cancer cells. SRE enhanced the ALP activity of pre-osteoblast MC3T3-E1 cells after 72 h in culture and slightly enhanced mineralization at 14

Transmissible spongiform encephalopathies (TSE) are characterized by the misfolding of the host encoded prion protein (PrP(C)) into a pathogenic isoform (PrP(Sc)) which leads to the accumulation of β-sheet-rich fibrils and subsequent loss of neurons and synaptic functions. Although many compounds have been identified which inhibit accumulation or dissolve fibrils and aggregates in vitro there is no therapeutic treatment to stop these progressive neurodegenerative diseases. Here we describe the effects of the traditional medicinal herb Scutellaria lateriflora (S. lateriflora) and its natural co
