Monitoring the kinetics of the pH-driven transition of the anthrax toxin prepore to the pore by biolayer interferometry and surface plasmon resonance.

Domain 2 of the anthrax protective antigen (PA) prepore heptamer unfolds and refolds during endosome acidification to generate an extended 100 Å β barrel pore that inserts into the endosomal membrane. The PA pore facilitates the pH-dependent unfolding and translocation of bound toxin enzymic components, lethal factor (LF) and/or edema factor, from the endosome to the cytoplasm. We constructed immobilized complexes of the prepore with the PA-binding domain of LF (LFN) to monitor the real-time prepore to pore kinetic transition using surface plasmon resonance and biolayer interferometry (BLI). Unified variational framework for simultaneous correction of non-uniform sampling and diffraction-induced slope errors in coherence scanning interferometry.

Coherence scanning interferometry (CSI) enables non-contact surface measurement with high axial resolution, but its accuracy is degraded in practical complex-surface metrology by two coupled effects: vibration-induced non-uniform axial sampling and diffraction-induced slope/edge errors. Axial-position sensing can provide frame-wise scanning positions, but it does not correct the optical-transfer-function degradation caused by finite-aperture imaging. Conversely, conventional inverse filtering compensates for diffraction-related attenuation but usually assumes uniform axial sampling. This paper

High-Sensitivity Cell-Based Plasmonic Biolayer Interferometry with a Two-Tiered Enhancement for Clinical Autoimmunity Surveillance of Pemphigus Vulgaris.

Pemphigus vulgaris (PV) is a chronic, often fatal autoimmune blistering disorder that affects the skin and body-wide mucous membranes, imperatively urging for accurate detection of characteristic autoantibodies. To overcome the sensitivity constraint, we report here a serological avidity test for complex PV autoantibodies, relying on cell-based plasmonic biolayer interferometry (cpBLI) with two-tiered enhancement of light interference by gold nanostars (AuNSs). High-surface-area, clean-surface AuNSs with abundant tips and grooves can enhance interference patterns and promote intrinsic and extr

Tear film interferometry assessment after intense pulsed light in dry eye disease: A randomized, single masked, sham-controlled study.

BACKGROUND: Inadequate meibomian glands (MGs) secretion can lead to dry eye signs and symptoms. Tear film lipid layer (TFLL) secreted by MGs protects and prevents rapid evaporation of tear film. The purpose was to assess TFLL alteration and function in patients with evaporative dry eye (EDE) using tear interferometry after optimal pulse light technology (OPT) intense pulsed light (IPL). METHODS: This prospective randomized examiner-masked sham- controlled study included 86 participants (142 eyes) with DED. IPL or sham procedure was performed on day 0, 21, and 42. A biolayer interferometry-based enzyme-linked aptamer sorbent assay for real-time and highly sensitive detection of PDGF-BB.

Accurate, fast and sensitive detection of disease-specific protein biomarkers, especially in blood, urine, or other bodily fluids, is an important approach to achieve early disease diagnosis. Platelet-derived growth factor-BB (PDGF-BB), a widely used biomarker, is involved in a substantial number of serious diseases, such as hepatic fibrosis, atherosclerosis, age-related macular degeneration and diabetic eye disease and is often over-expressed in human malignant tumors. Therefore, the development of sensitive and specific detection methods for PDGF-BB is of great importance for the early diagn

Estimation of ocular rigidity based on measurement of pulse amplitude using pneumotonometry and fundus pulse using laser interferometry in glaucoma.

PURPOSE: There is evidence from theoretical models and animal studies that the biomechanical properties of the optic nerve head and the sclera play a role in the pathophysiology of glaucoma. There are, however, only a few data available that demonstrate such biomechanical alterations in vivo. In this study, the hypothesis was that patients with primary open-angle glaucoma (POAG) have an abnormal ocular structural stiffness based on measurements of intraocular pressure amplitude and ocular fundus pulsation amplitude (FPA). METHODS: Seventy patients with POAG and 70 healthy control subjects mat

Expanding Biolayer Interferometry Applications: Enhanced Accuracy, Precision, and Sensitivity in Residual Biomolecule Detection and Quantitation of Bispecifics and AAV Viral Particles.

Biolayer Interferometry (BLI) has traditionally been used for characterization of protein-protein interactions (PPI) with proteins, such as antibodies and their antigens, through kinetic and quantitation assays. Limitations, for example in sensitivity and the availability of established assay formats, have restricted its adoption across other analytical applications. This article highlights three case studies which demonstrate the expansion of BLI into novel applications, spanning the areas of protein detection and viral vector characterization. The first case study details the use of a multi-

In vitro optical characterization of the RayOne Galaxy spiral extended depth-of-focus intraocular lens using high-resolution Mach-Zehnder interferometry.

Spiral optics have been proposed to extend depth of focus through controlled phase modulation. We characterized the RayOne Galaxy spiral intraocular lens using high-resolution Mach-Zehnder interferometry and a simulated pseudophakic Arizona eye model. Wavefronts were decomposed into Zernike polynomials and compared with monofocal, trifocal, and spherical-aberration-based extended-depth-of-focus (EDOF) lenses. The Galaxy exhibited a smooth EDOF profile with a moderate reduction in contrast rendering. Symmetry analysis showed that radially symmetric aberrations primarily generated the EDOF effec

Effects from metal ion in tumor endothelial marker 8 and anthrax protective antigen: BioLayer Interferometry experiment and molecular dynamics simulation study.

One of the anthrax receptors, tumor endothelial marker 8 (TEM8), is reported to be a potential anticancer target due to its over-expression during tumor angiogenesis. To extend our BioLayer Interferometry study in PA-TEM8 binding, we present a computational approach to reveal the role of an integral metal ion on receptor structure and binding thermodynamics. We estimated the interaction energy between PA and TEM8 using computer simulation. Consistent with experimental study, computational results indicate the metal ion in TEM8 contributes significantly to the binding affinity, and PA-TEM8 bind

Developing the Readout Electronics for a Custom 64 × 64 SPAD Array: From Single-Board Prototyping to FPGA Implementation Toward Stellar Intensity Interferometry.

Single-photon avalanche diode (SPAD) arrays enable photon-starved applications, including time-of-flight imaging and stellar intensity interferometry. Their astronomical use remains scarcely explored, as the bottleneck is usually not detection but rather acquisition electronics for high-rate event streams. This work presents a modular, event-driven acquisition system for a 64 × 64 SPAD array within the La Palma Quantum Interferometer (LPQI) project, repurposing a LiDAR detector for multi-telescope interferometry. Two stages are used: a Raspberry Pi 5 with a custom board for validation, and an
