Hyaluronic acid-based dissolving microneedles for enhanced cutaneous delivery of 3-Acetyl-11-keto-β-boswellic acid in rheumatoid arthritis: In vitro, ex vivo, and preclinical investigation.

Rheumatoid arthritis (RA) is a chronic inflammatory disorder characterized by synovial inflammation, joint destruction, and systemic complications. The management of RA often involves the use of anti-inflammatory and anti-arthritic agents. One natural compound 3-Acetyl-11-keto-β-boswellic acid (AKBA) showed promising therapeutic potential in RA conditions, however, the clinical application of AKBA is hindered by its poor solubility and permeability. Therefore, we aimed to develop an effective AKBA delivery system for the application in RA. We developed a novel hyaluronic acid-based dissolvable

Inhibition of heme detoxification pathway in malaria parasite by 3-hydroxy-11-keto-β-boswellic acid isolated from Boswellia serrata.

Malaria eradication is still a major global health problem in developing countries, which has been of more concern ever since the malaria parasite has developed resistance against frontline antimalarial drugs. Historical evidence proves that the plants possess a major resource for the development of novel anti-malarial drugs. In the present study, the bioactivity guided fractionation of the oleogum-resin of Boswellia serrata Roxb. yielded the optimum activity in the ethyl acetate fraction with an IC50 of 22 ± 3.9 μg/mL and 26.5 ± 4.5 μg/mL against chloroquine sensitive (NF54) and resistant (K1

3-Acetyl-11-keto-β-boswellic acid attenuated oxidative glutamate toxicity in neuron-like cell lines by apoptosis inhibition.

Источник пометил публикацию как отозванную

3-Acetyl-11-keto-β-boswellic acid (AKBA), a pentacyclic triterpenic acid present in gum resin of Boswellia serrata, has been found to possess antioxidant and neuroprotective properties. In this study, we aimed to examine protective properties of AKBA against glutamate-induced neuronal injury. To investigate the effects of AKBA (2.5-10 µM) on glutamate injury in neuron-like cells PC12 and N2a, two treatment regimens (incubation for 2 or 0 hours before glutamate exposure) were used. Then, the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide method was used to determine viability of t

Alteration in Amino Acid Metabolism After Isocaloric, Energy-Restricted Ketogenic Diet in Women with Overweight and Obesity: Randomized KETO-MINOX Trial.

Background/Objectives: Circulating amino acid concentrations and their excretion can provide insights into dietary protein intake and metabolism. Alterations in amino acid homeostasis occur in various disorders due to nutritional imbalances or metabolic changes, including obesity. A ketogenic diet (KD) has gained popularity for weight management; however, its metabolic effects are not fully known. Therefore, the aim of this study was to evaluate the effect of an eight-week, energy-restricted Mediterranean-type KD on the amino acid metabolism in women with overweight and class I obesity. Iron(II)-α-keto acid complexes of tridentate ligands on gold nanoparticles: the effect of ligand geometry and immobilization on their dioxygen-dependent reactivity.

Two mononuclear nonheme iron(II)-benzoylformate (BF) complexes [(6Me2-Me-BPA)Fe(BF)](ClO4) (1a) and [(6Me3-TPMM)Fe(BF)](ClO4) (1b) of tridentate nitrogen donor ligands, bis((6-methylpyridin-2-yl)methyl)(N-methyl)amine (6Me2-Me-BPA) and tris(2-(6-methyl)pyridyl)methoxymethane (6Me3-TPMM), were isolated and characterized. The structural characterization of iron(II)-chloro complexes indicates that the ligand 6Me2-Me-BPA binds to the iron(II) centre in a meridional fashion, whereas 6Me3-TPMM behaves as a facial ligand. Both the ligands were functionalized with terminal thiol for immobilization on

alpha-keto-beta-methyl-n-valeric acid diminishes reactive oxygen species and alters endoplasmic reticulum Ca(2+) stores.

Mitochondrial dysfunction and oxidative stress occur in neurodegenerative diseases. Other results show that bombesin-releasable calcium stores (BRCS) from the endoplasmic reticulum (ER) are exaggerated in fibroblasts from patients with Alzheimer's disease (AD) compared with controls and in fibroblasts from a young control treated with H(2)O(2). We hypothesize that alterations in oxidative stress underlie the exaggeration in BRCS in AD, and that appropriate antioxidants may be useful in treating this abnormality. Two indicators of different oxidant species were used to determine the effects of

Efficient Production of 2-Keto-l-Gulonic Acid via One-Step Fermentation Using Gluconobacter oxydans WTF0512 and Ketogulonicigenium vulgare WTF0114.

Currently, the main method for producing the vitamin C precursor 2-keto-l-gulonic acid (2-KLG) is a two-step fermentation process, in which secondary sterilization and fermentation processes result in higher costs and energy consumption. Consequently, the development of a one-step fermentation process is seen as a more desirable approach for 2-KLG production. In this study, we used Gluconobacter oxydans WTF0512 and Ketogulonicigenium vulgare WTF0114 as co-cultured strains for the production of 2-KLG from d-sorbitol via one-step fermentation. The fermentation behaviors of G. oxydans WTF0512 and

Acetyl-11-keto-β-boswellia acid attenuates Ti particle-induced osteoblastic oxidative stress and osteolysis through the Foxo3 signaling pathway.

Oxidative stress injury in osteoblasts is one of the leading causes of periprosthetic osteolysis (PPOL). Acetyl-11-keto-β-boswellia acid (AKBA) has been used as an antioxidant in the treatment of various diseases, but its antioxidant mechanism in osteolysis has yet to be elucidated. In this study, a mouse cranial osteolysis model was constructed, and MC3T3-E1 cells and bone marrow mesenchymal stem cells (BMSCs) were cultured in vitro. Western blotting and immunofluorescence staining revealed that titanium (Ti) particles aggravated osteoblast oxidative stress injury and apoptosis. Ti particles

Increased valinomycin production in mutants of Streptomyces sp. M10 defective in bafilomycin biosynthesis and branched-chain α-keto acid dehydrogenase complex expression.

Streptomyces sp. M10 is a valinomycin-producing bacterial strain that shows potent bioactivity against Botrytis blight of cucumber plants. During studies to increase the yield of valinomycin (a cyclododecadepsipeptide) in strain M10, additional antifungal metabolites, including bafilomycin derivatives (macrolide antibiotics), were identified. To examine the effect of bafilomycin biosynthesis on valinomycin production, the bafilomycin biosynthetic gene cluster was cloned from the genome of strain M10, as were two branched-chain α-keto acid dehydrogenase (BCDH) gene clusters related to precursor

Two novel mutations in the BCKDK (branched-chain keto-acid dehydrogenase kinase) gene are responsible for a neurobehavioral deficit in two pediatric unrelated patients.

Inactivating mutations in the BCKDK gene, which codes for the kinase responsible for the negative regulation of the branched-chain α-keto acid dehydrogenase complex (BCKD), have recently been associated with a form of autism in three families. In this work, two novel exonic BCKDK mutations, c.520C>G/p.R174G and c.1166T>C/p.L389P, were identified at the homozygous state in two unrelated children with persistently reduced body fluid levels of branched-chain amino acids (BCAAs), developmental delay, microcephaly, and neurobehavioral abnormalities. Functional analysis of the mutations confirmed th

Regulation of hepatic branched-chain alpha-keto acid dehydrogenase kinase in a rat model for type 2 diabetes mellitus at different stages of the disease.

Branched-chain alpha-keto acid dehydrogenase (BCKDH) kinase (BDK) is responsible for the regulation of BCKDH complex, which is the rate-limiting enzyme in the catabolism of branched-chain amino acids (BCAAs). In the present study, we investigated the expression and activity of hepatic BDK in spontaneous type 2 diabetes using hyperinsulinemic Zucker diabetic fatty rats aged 9weeks and hyperglycemic, but not hyperinsulinemic rats aged 18weeks. The abundance of hepatic BDK mRNA and total BDK protein did not correlate with changes in serum insulin concentrations. On the other hand, the amount of B

Molecular structure of 3-O-acetyl-11-keto-β-boswellic acid and its ʟ-alanine conjugate (AKBA-Ala) and their impact on lipoxygenases.

3-O-Acetyl-11-keto-β-boswellic acid (AKBA) is an anti-inflammatory constituent of frankincense. Its potency is attributed to its ability to interact with enzymes in the inflammation cascade, such as lipoxygenase and cyclooxygenase, to decrease formation of pro-inflammatory products while increasing the production of pro-resolving mediators. However, the availability of AKBA in vivo may be limited by factors such as poor solubility and protein binding in serum. A prodrug approach, wherein AKBA is conjugated to amino acids through its carboxylate moiety, could be a suitable way to improve solubi

Boswellia serrata extract with low 3O-acetyl-11-keto-β-boswellic acid-content causes efficient lipid mediator class switch.

BACKGROUND: Extracts of the gum resin from Boswellia (B.) serrata Roxb. (Burseraceae), termed frankincense, are popular natural remedies for treatment of inflammatory disorders, and clinical trials confirm the anti-inflammatory efficacy, especially in osteoarthritis. The pentacyclic triterpenes boswellic acids are exclusive to frankincense, where 3-O-acetyl-11-keto-β-boswellic (AKBA) is considered as the main active principle. AKBA allosterically modulates lipoxygenases (LOX) in lipid mediator (LM) biosynthesis. This LOX modulation leads to suppression of pro-inflammatory leukotrienes while in

Boosting cutaneous penetration of 3-acetyl-11-keto-β-boswellic acid by glycosaminoglycan-based polysaccharide coated proglycosomes for rheumatoid arthritis therapy.

3-Acetyl-11-keto-β-boswellic acid (AKBA), a plant-derived anti-inflammatory agent, has shown therapeutic potential in the management of rheumatoid arthritis (RA). However, its clinical utility is limited by poor solubility and low bioavailability. To address these challenges, the present study aimed to develop and evaluate surface-engineered proglycosomes (PGS) coated with glycosaminoglycan-based biological macromolecules, hyaluronic acid (HA), and chondroitin sulfate (CS) as (HA-AKBA-PGS and CS-AKBA-PGS) for targeted dermal delivery of AKBA. Molecular basis of intermittent maple syrup urine disease: novel mutations in the E2 gene of the branched-chain alpha-keto acid dehydrogenase complex.

The E2 gene of the branched-chain alpha-keto acid dehydrogenase (BCKDH) complex was studied at the molecular level in three patients with intermittent maple syrup urine disease (MSUD). All three patients had higher BCKDH activity than did those with the classical phenotype. In the first patient, a single base substitution from A to G in intron 8 created a new 5' splice site and caused an insertion of 126 nucleotides between exons 8 and 9 by activating an upstream cryptic 3' splice site in the same intron. Characterization of a fourth type of 2-keto acid-oxidizing enzyme from a hyperthermophilic archaeon: 2-ketoglutarate ferredoxin oxidoreductase from Thermococcus litoralis.

Thermococcus litoralis is a strictly anaerobic archaeon (archaebacterium) that grows at temperatures up to 98 degrees C by fermenting peptides. It is known to contain three distinct ferredoxin-dependent, 2-keto acid oxidoreductases, which use pyruvate, aromatic 2-keto acids such as indolepyruvate, or branched-chain 2-keto acids such as 2-ketoisovalerate, as their primary substrates. We show here that T. litoralis also contains a fourth member of this family of enzymes, 2-ketoglutarate ferredoxin oxidoreductase (KGOR). In the presence of coenzyme A, KGOR catalyzes the oxidative decarboxylation
