Demonstration of myenteric plexus abnormalities in genetic diseases by a microdissection technique: preliminary studies.

Eighty-eight specimens of esophagus, small intestine, or colon from 45 patients, predominantly infants and children, with 30 different genetic diseases were analyzed by a microdissection technique for the following abnormalities of the Auerbach (myenteric) plexus: (1) abnormality of the pattern of the nervous network of the plexus, (2) abnormal fraction of neural tissue in the plane of the plexus, (3) abnormal size or appearance of the cytoplasm of the neurons of the plexus, and (4) abnormal number of neurons in the ganglia of the plexus. Seven of 8 specimens of esophagus from patients with ne

Intestinal plexuses in Crohn's disease and ulcerative colitis in children: pathologic and microdissection studies.

The relative fraction of neural tissue in the plane of the myenteric (Auerbach) plexus (MEP) of ileum and colon of patients with Crohn's disease and ulcerative colitis was analyzed by point-count morphometric analysis of microdissected flat-mount ("circuit diagram") preparations of the plexus. Increase in the relative fraction of neural tissue (myenteric plexus hyperplasia) was seen in the MEP of some patients in both ileum and colon in both diseases. The frequency of ulceration or inflammation extending into the bowel wall deep enough to injure the various levels of the intestinal plexus syst

Alpha 2-adrenoceptor stimulation and cellular cAMP levels in microdissected rat glomeruli.

A functional role for the numerically predominant glomerular alpha 2-adrenoceptors is unknown. In other tissues, activation of alpha 2-adrenoceptors inhibits adenylate cyclase activity. We therefore examined the effect of alpha 2-adrenoceptor stimulation with (-)-epinephrine (E) on the cellular cAMP concentration in glomeruli isolated by microdissection. Parathyroid hormone (1-34 PTH), prostaglandin E2 (PGE2), histamine, serotonin, or adenosine, in the presence of 3-isobutyl-1-methylxanthine (phosphodiesterase inhibitor) and propranolol, was used to activate adenylate cyclase in single intact

High-Resolution Laser Microdissection to Investigate Transcriptional States in Formalin-Fixed Paraffin-Embedded Samples.

The advent of spatially resolved transcriptomics has revolutionized our understanding of tissue heterogeneity. While advanced spatial transcriptomics (ST) technologies offer unparalleled and unbiased, whole-slide molecular mapping, laser microdissection (LMD) remains a highly valuable and often superior approach for specific research questions, particularly when coupled with RNA sequencing (RNA-seq). LMD provides high spatial resolution through precise morphological annotation, allowing for the isolation of defined cell clusters or even rare populations from complex tissues. This enables deepe

Immuno-laser capture microdissection of perfusion-fixed mouse brain tissue coupled to RNA-seq.

BACKGROUND: Laser capture microdissection (LCM), when combined with immunostaining (immuno-LCM) and coupled to RNA profiling, provides a means for in situ transcriptomic interrogation of complex tissues. However, preserving RNA integrity through the multiple harsh steps of immuno-LCM has proved challenging, greatly limiting the potential for high-resolution spatial analysis of global gene expression. NEW METHOD: Here, we describe a protocol whereby perfusion fixation and subsequent ex vivo post-fixation of mouse brain with paraformaldehyde, followed by protease digestion of immuno-LCM-acquire

Robust Acquisition of Spatial Transcriptional Programs in Tissues With Immunofluorescence-Guided Laser Capture Microdissection.

The functioning of tissues is fundamentally dependent upon not only the phenotypes of the constituent cells but also their spatial organization in the tissue, as local interactions precipitate intra-cellular events that often lead to changes in expression. However, our understanding of these processes in tissues, whether healthy or diseased, is limited at present owing to the difficulty in acquiring comprehensive transcriptional programs of spatially- and phenotypically-defined cells in situ. Here we present a robust method based on immunofluorescence-guided laser capture microdissection (immu

Insights into olfactory ensheathing cell development from a laser-microdissection and transcriptome-profiling approach.

Olfactory ensheathing cells (OECs) are neural crest-derived glia that ensheath bundles of olfactory axons from their peripheral origins in the olfactory epithelium to their central targets in the olfactory bulb. We took an unbiased laser microdissection and differential RNA-seq approach, validated by in situ hybridization, to identify candidate molecular mechanisms underlying mouse OEC development and differences with the neural crest-derived Schwann cells developing on other peripheral nerves. We identified 25 novel markers for developing OECs in the olfactory mucosa and/or the olfactory nerv

Purification of diseased cells from Barrett's esophagus and related lesions by laser capture microdissection.

Barrett's esophageal adenocarcinoma (BEAC) arises from Barrett's esophagus (BE), a premalignant lesion caused by acid reflux (heartburn). Although the cancer is uncommon, its incidence is rapidly rising in western countries. Like most other cancers, BEAC cells also have elevated telomerase activity which maintains telomere length and supports continued proliferation of these cells. It is not clear if telomerase is activated early at premalignant (BE) stage, because reports of telomerase activity in Barrett's and normal esophagi have been controversial. We have shown that detection of telomeras

Selective capture of endothelial and perivascular cells from brain microvessels using laser capture microdissection.

Laser capture microdissection (LCM) of the major cell types comprising brain microvessels offers a powerful technology to explore the molecular basis of the blood-brain barrier in health and disease. However, the ability to selectively retrieve endothelial or perivascular cells, without cross-contamination from the other, has proven difficult. Additionally, histochemical methods previously described for use with LCM have not allowed for identification of all the different size branches of the microvascular tree. Here, we describe a double immunostaining method, combining bright-field and fluor

Compartment-specific quantitative gene expression analysis after laser microdissection from archival renal allograft biopsies.

BACKGROUND: Various immunological and non-immunological pathomechanisms are responsible for the cellular damage in renal allografts. Since the kidney is an anatomically complex organ with functional and morphological heterogeneous compartments (interstitium, tubuli, vessels, glomeruli), the local response to injury maybe variable, therefore, the identification of local pathomechanisms is important. AIM: To elucidate any discrepancies in quantitative mRNA expression profiles between a total specimen analysis and a cell-specific evaluation after laser microdissection. METHODS: Real-time RT-PCR

Intracellular expression profiling by laser capture microdissection: three novel components of the neuromuscular junction.

Amiloride-sensitive sodium entry, via the epithelial sodium channel (ENaC), is the rate-limiting step for Na+ absorption in kidney collecting ducts, and epidermal growth factor (EGF) inhibits Na+ transport and ENaC expression. A pathognomonic feature of polycystic kidney disease (PKD) is EGF receptor mislocalization to the apical plasma membrane and EGF/EGF receptor axis overactivity. Immunohistochemical and biochemical analysis revealed mislocalization of EGF receptor and excessive activation of the p42/44 extracellular signal-regulated protein kinase pathway (ERK1/2) in kidneys from cystic m

Chronic inflammatory ear disease and cholesteatoma: creation of auxiliary attic aeration pathways by microdissection.

HYPOTHESIS: The attic compartments, except for Prussak's space, are aerated through the tympanic isthmus. The aim of this study was to develop aeration pathways that would bypass the isthmus in surgery for chronic inflammatory ear disease and cholesteatoma. BACKGROUND: Microdissection of the epitympanum has shown that the anterior attic and the supratubal recess are separated by the tensor fold, the excision of which creates a large new aeration pathway. METHODS: Earlier surgical experience was reexamined as to the access to the tensor fold. Twenty temporal bones were dissected to create cli

Characterization of familial partial 10p trisomy by chromosomal microdissection, FISH, and microsatellite dosage analysis.

Unbalanced translocations are a frequent cause of multiple congenital anomalies in children. Translocations as small as 2-5 Mb of DNA are detectable by G-banding under optimal conditions. Some of these small translocations are visible but cannot be characterized cytogenetically due to the lack of characteristic banding on Giemsa preparations. We have combined chromosomal microdissection and fluorescence in situ hybridization (FISH) to identify the origin of a small translocated segment in three members of a family with a derivative chromosome 9 and multiple anomalies, including several ophthal

Outcomes of microdissection testicular sperm extraction in azoospermic men following chemotherapy: A cross-sectional study.

BACKGROUND: The use of chemotherapy agents can increase the likelihood of azoospermia in men. Today, the application of micro-dissection testicular sperm extraction (micro-TESE) and subsequent intracytoplasmic sperm injection (ICSI) is recommended for sperm retrieval in such affected men. OBJECTIVE: We aimed to report our experiences with the success rate of the protocol of micro-TESE in maintaining sperm restoration in men undergoing chemotherapy protocols for different cancers. MATERIALS AND METHODS: In this cross-sectional study, data of 33 men who were scheduled for micro-TESE and subseq

Neurovascular compression from a tortuous vertebrobasilar system. Technical note on surgical anatomy and cisternal microdissection.

BACKGROUND: Microvascular decompression (MVD) for trigeminal neuralgia (TGN) typically entails mobilisation of the superior cerebellar artery or adjacent veins. Rarely, symptomatic compression arises from a tortuous vertebrobasilar system. These cases are challenging due to eloquence and distorted microsurgical anatomy in cerebellopontine angle (CPA) often needing more complex maneuvers like vessel transposition rather than conventional interposition technique. This technical note discusses MVD technical nuances for TGN from a tortuous vertebrobasilar system. METHOD: Standard retrosigmoid cra

True Schistosoma mansoni eggs-cretome revealed by laser microdissection of infected mouse liver and intestine.

INTRODUCTION: Schistosoma mansoni eggs are the central drivers of schistosomiasis pathology and transmission, yet the proteins they release within host tissues remain poorly defined due to limitations of conventional in vitro approaches. Here, we provide the first in situ characterization of egg-derived proteins present in host tissues. METHODS: Using laser microdissection, we selectively isolated periovular granuloma microenvironments from the livers and small intestines of infected mice during the acute and chronic phases of infection, followed by high-resolution mass spectrometry. Optimized protocol to preserve RNA integrity for laser capture microdissection of bovine mammary epithelial cells.

Laser capture microdissection (LCM) is a popular technique for isolating specific cell types from tissues for medical research. However, the LCM procedure for isolating bovine mammary epithelial cells (MEC) for high-throughput transcriptome profiling has been lacking. The quality and quantity of RNA in LCM samples can be significantly affected by tissue treatment, time for dissection, and the total area of cells dissected. The objective of this study was to optimize the procedures for isolating bovine MEC from mammary tissues for consistent isolation of intact RNA suitable for downstream high-
