The exquisite preservation of maxillary and mandibular fragments of Seymouria has allowed us to examine for the first time in detail the dental anatomy and patterns of development in this stem amniote. The results obtained through histological examination show that Seymouria has pleurodont implantation with ankylosis of the tooth to the labial side of the jawbone. The dentary and maxillary teeth exhibit similar dental characteristics, such as the attachment bone (alveolar bone) and cementum rising above the jawbone on the base of the tooth, and smooth carinae extending lingually toward the too

In the course of an evolutionary and developmental study on the dermal skeleton, our attention was drawn to the existence of denticles located outside the oral cavity in the atheriniform species Atherion elymus. These denticles, attached to the surface of most dermal bones of the head, are especially numerous on the snout, chin and the undersides of the lower region of the head, where they are aligned forming a crenulated keel. Using light, scanning and transmission electron microscopy, we clearly demonstrate the dental (vs bony) nature of these denticles. They are small, conical elements most

The epidermic growth factor (EGF) inhibits the morphogenesis and cellular differentiation of dental tissues during embryogenesis. These effects are modulated according to the stage of tooth development. Thus, the cupula stage seems markedly affected contrary to the bell stage which continues to evolve eve in the presence of EGF. Distribution of EGF receptors is altered during the first stages or the morphogenesis and this is specific for each stage of dental development. Finally, some rare pathological processes, related to the EGF level, are particularly characterized by a very early growth o

Tooth development and replacement in fetal and adult viviparous caecilians (Amphibia: Gymnophiona) are described and analyzed according to current theories of tooth succession. The fetal dentition differs from that of the adult in morphology, position, and function. Teeth are used by fetuses to scrape the oviducal epithelium, thus stimulating the secretion of a nutrient substance. Fetal dentitions vary in morphology and position in different species. The ontogeny of teeth of several species is described and the patterns of addition of loci and of replacement are analyzed. Loci are added both p

INTRODUCTION: Supernumerary teeth in cases of cleft lip and palate do not result from the division of normal germs before the formation of hard tissue. Deciduous and permanent teeth odontogenesis begins after the face has formed, either with or without the cleft.
DISCUSSION: The most acceptable hypothesis to enable understanding of the presence of supernumerary teeth on one or both sides of the cleft palate is hyperactivity of the dental lamina in its walls. This hyperactivity, with the formation of more tooth germs, must be attributed to mediators and genes related to tooth formation, under

Fibroblast growth factors (FGFs) are diffusible polypeptides released by a variety of cell types. FGF8 subfamily members regulate embryonic development processes through controlling progenitor cell growth and differentiation, and are also functional in adults in tissue repair to maintain tissue homeostasis. FGF8 family members exhibit unique binding affinities with FGF receptors and tissue distribution patterns. Increasing evidence suggests that, by regulating multiple cellular signaling pathways, alterations in the FGF8 subfamily are involved in craniofacial development, odontogenesis, tongue

Sequential and reciprocal epithelial-mesenchymal interactions, essential throughout such aspects of tooth morphogenesis as patterning, size and number of teeth, involves a well-ordered series of inductive and permissive signals that exert global control over cell proliferation, differentiation and organogenesis. In particular, growth factors, transcription factors and their corresponding receptors, as well as other soluble morphogens, make up a regulatory network at the molecular level that synergistically or antagonistically controls intra-/inter-cellular signal transduction during odontogene

Williams syndrome is a rare congenital disorder involving the cardiovascular system, mental retardation, distinctive facial features, and tooth anomalies. It is caused by the heterozygous deletion of approximately 1.6 Mb encompassing 28 genes on human chromosome 7q11.23. It has been suggested that the genes responsible for craniofacial anomalies are located in the telomeric end region, which harbors three members of the TFII-I gene family (Tassabehji et al. [2005] Science 310:1184). To recognize potential candidate genes for the tooth anomalies in Williams syndrome, we carried out comparative

Mineralization is a critical process in the formation of hard tissues such as bones and teeth, yet the cellular mechanisms underlying this process remain incompletely understood. To address this, we constructed a comprehensive single-cell atlas of tooth development, integrating data from 261,929 cells across 15 projects and encompassing both odontogenesis and amelogenesis. We developed a novel algorithm, TrajDTW, to identify genes with concordant trajectory dynamics across these large-scale datasets, allowing us to detect robust developmental signals. To define a shared mineralization trajecto

The pesticide Dichlorodiphenyltrichloroethane (DDT) and its degradation by-products (Dichlorodiphenyldichloroethane-DDD and dichlorodiphenyldichloroethylene-DDE) are health concerns. This study aimed to investigate the impact of multigenerational exposure to traces of DDD and DDE on odontogenesis Wistar rats were allocated in 4 groups over 2 generations (F1 and F2). Water was provided ad libitum throughout the animal's life groups: control - natural water; DDD - 0.0147 µM DDD; DDE - 0.0056 µM DDE; and DDD+DDE - mixture of the two solutions. They were euthanized and the heads removed for comput

Tooth root development involves intricate spatiotemporal cellular dynamics and molecular regulation. The initiation of Hertwig's epithelial root sheath (HERS) induces odontoblast differentiation and the subsequent radicular dentin deposition. Precisely controlled signaling pathways modulate the behaviors of HERS and the fates of dental mesenchymal stem cells (DMSCs). Disruptions in these pathways lead to defects in root development, such as shortened roots and furcation abnormalities. Advances in dental stem cells, biomaterials, and bioprinting show immense promise for bioengineered tooth root

BACKGROUND: Tooth development is highly regulated in mammals and it is regulated by networks of signaling pathways (e. g. Tnf, Wnt, Shh, Fgf and Bmp) whose activities are controlled by the balance between ligands, activators, inhibitors and receptors. The members of the R-spondin family are known as activators of Wnt signaling, and Lgr4, Lgr5, and Lgr6 have been identified as receptors for R-spondins. The role of R-spondin/Lgr signaling in tooth development, however, remains unclear.
RESULTS: We first carried out comparative in situ hybridization analysis of R-spondins and Lgrs, and identifie

The anterior alveolar ridge of the maxilla is a frequent site for pathology, most of it involving underlying bone and readily observable radiologically. Although much of this disease presents as well-defined unilocular radiolucencies arising adjacent to the apices of non-vital anterior teeth, thus being periapical radiolucencies of inflammatory origin (PRIO), a minority reflects more serious disease such as neoplasia. The most serious of the latter are malignancies, most frequently squamous cell carcinomas (SCCs). Although the majority of SCCs arise from the oral or nasal mucosa and secondaril

Introduction. "Dens invaginatus" is a dental anomaly which originates from the invagination of the ameloblastic epithelium into the lingual surface of the dental crown during the odontogenesis. It can cause early pulpal necrosis, abscesses, retention or dislocation of contiguous elements, cysts, and internal resorptions. It normally affects the upper lateral incisors. In the following study the authors will discuss the etiology, the physiopathology, and the surgical-orthodontic management of a rare case of impacted canine associated with dens invaginatus and follicular cyst, with the aim of hi

BACKGROUND: Hyperdontia is a disorder of odontogenesis characterised by excess teeth. Many complications can be associated with supernumerary teeth, such as crowding, tooth displacement, diastema, deep caries, retention or impaction, delayed eruption or ectopic eruption of adjacent teeth, aesthetic problems, neuralgic manifestations, root resorption of adjacent teeth, and dentigerous cyst formation with significant bone destruction. Treatment of hyperdontia depends on the area and number of supernumerary teeth, and also on the presence of pathologic processes that can affect them and/or the te

Substantial progress has been made regarding the molecular etiology of the dentin diseases dentinogenesis imperfecta types II and III, and dentin dysplasia type II. Genetic linkage studies have identified the critical loci for these diseases on human chromosome 4q21. Located within an overlapping segment of these disease loci is a dentin/bone gene cluster that includes osteopontin, bone sialoprotein, dentin matrix protein 1, dentin sialophosphoprotein, and a new gene MEPE also known as OF45, renamed osteoregulin. In this paper, we report the location of MEPE/OF45 in relationship to the other m

BACKGROUND: Given the susceptibility of developing tissues to drugs, even small doses of anticancer drugs may affect odontogenesis. Although any toxic effect is transient, the treatment regimens are based on repeated drug administration.
OBJECTIVES: The study aimed to establish the impact of antineoplastic therapy on the occurrence of longterm adverse dental effects in a dose-dependent manner in young survivors treated for cancer before 10 years of age.
MATERIAL AND METHODS: In total, 37 cancer survivors treated with antineoplastic therapy before 10 years of age underwent a dental examinatio
