Experimental reproduction of malabsorption syndrome with different combinations of reovirus, Escherichia coli, and treated homogenates obtained from broilers.

Attempts to reproduce malabsorption syndrome (MAS) by oral inoculation with several different combinations including intestinal homogenate, reovirus, and hemolytic Escherichia coli obtained from MAS-affected chickens and intestinal homogenate from healthy chickens (healthy homogenate) were performed in 1-day-old specific-pathogen-free (SPF) broilers. The MAS homogenate, serving as a positive control, induced weight gain depression and intestinal lesions such as cystic crypts of Lieberkuhn, villus atrophy, and lymphoid and/or granulocytic infiltration. The healthy homogenate, the formalin-treat

Effect of bacteria on the inactivation and adsorption on clay minerals of reovirus.

This investigation studied the antiviral activity of, and the utilization of viruses as substrates by, bacteria. Reovirus type 3 and bacterial species representative of those endemic to sewage, aquatic, and terrestrial habitats were used in the model systems. Culture supernatants from Bacillus subtilis maintained for 5 days in a minimal salts medium displayed antiviral activity, but supernatants from Escherichia coli or Serratia marcescens did not. Both live and toluene-killed cells reduced the inactivation of reovirus during 4 days of incubation at 23 +/- 2 degrees C. This protective effect w

A retrospective analysis of cystic enteropathy in Alabama broilers.

Runting-stunting syndrome (RSS) is an enteric disease of young chickens with complex aetiopathogenesis. It is often linked to the isolation of multiple virus species and histologically characterized by cystic enteropathy. We investigated the correlation between the detection frequency of avian reovirus, astrovirus, parvovirus, and rotavirus A and the presence of microscopic lesions consistent with cystic enteropathy in 123 broiler chicken submissions to the Alabama State Laboratory System (Auburn, AL, USA). Multi-viral detections were found to be predictive of cystic enteropathy with an increa

Serum metabolomic profiling reveals LysoPC/PC depletion as a potential biomarker to detect avian reoviral infection in neonatal broiler chickens.

INTRODUCTION: Avian reovirus (ARV) infection causes viral arthritis in broiler chickens, leading to lameness and substantial economic loss in poultry production. Current conventional diagnostic methods detect infection only after clinical signs appear, limiting the ability to prevent disease progression. Hence, this study utilized serum metabolomics to identify early biomarkers of ARV infection prior to tissue damage. METHODS: A total of 150 serum samples were collected from ARV-challenged broilers at 24, 48, and 72 h post-infection, along with uninfected controls. The samples underwent liqui

Wnt14 regulates the cellular inflammation induced by avian reovirus and interacts with the viral σB protein.

Avian reovirus (ARV) typically induces viral arthritis or tenosynovitis in chickens. The ARV structural protein σB plays a crucial role in viral replication and regulates cellular signaling pathways through interactions with host proteins. In this study, we demonstrate that both ARV infection and ARV σB protein can activate intracellular Wnt signaling pathways and induce inflammatory responses based on qPCR and Western blot analyses in HD11 cells. Interestingly, ARV infection can inhibit β-catenin ubiquitination and upregulate its protein expression levels. To further investigate the mechanism

Pathogenicity of avian reovirus variant in the immune organs of broiler chicks.

In recent years, the infection rate of avian reovirus (ARV) in chicks has been continuously increasing, seriously endangering the healthy development of China's chicken farming industry. However, the pathogenicity of this virus in immune organs has been less studied. In this study, an ARV strain (Chicken/XJ/China/2022/01/16) was isolated from arthritic broiler chicks in Xinjiang province and whole genome sequencing of the ARV strain was performed using next generation sequencing (NGS) technology, and sequence analysis showed that the strain was a type V variant of the ARV gene. In order to fur

Avian Reoviruses from Clinical Cases of Tenosynovitis: An Overview of Diagnostic Approaches and 10-Year Review of Isolations and Genetic Characterization.

Reoviral-induced tenosynovitis/viral arthritis is an economically significant disease of poultry. Affected birds present with lameness, unilateral or bilateral swollen hock joints or shanks, and/or reluctance to move. In severe cases, rupture of the gastrocnemius or digital flexor tendons may occur, and significant culling may be necessary. Historically, vaccination with a combination of modified live and inactivated vaccines has successfully controlled disease. Proper vaccination reduced vertical transmission and provided maternal-derived antibodies to progeny to protect against disease, at a

Identification of Coronaviruses, Paramyxoviruses, Reoviruses, and Rotaviruses among Bats in Nigeria.

Bats are often consumed by some ethnic groups in Nigeria despite association of bats with many important emerging viruses. More than 300 bats representing eight species were captured during 2010-2011 in eight locations of northern Nigeria. Available fecal swabs (n = 95) were screened for the presence of arenaviruses, CoVs, paramyxoviruses (PMVs), reoviruses, rhabdoviruses, and influenza viruses using generic reverse transcription-polymerase chain reaction assays. Here, we document the detection of CoVs, PMVs, reoviruses, and rotaviruses (RVs) in Nigerian bats. The Nigerian bat CoVs are grouped

Mixed infection with reovirus and Chlamydophila in a flock of budgerigars (Melopsittacus undulatus).

Eleven budgerigars (Melopsittacus undulatus) from a zoological collection presented at necropsy with emaciation and splenomegaly or hepatomegaly or both. Polymerase chain reaction assays performed on liver and spleen samples were positive for Chlamydophila psittaci in 2 of 3 birds tested, and histologic findings in 2 additional birds were compatible with chlamydiosis. The aviary was subsequently closed to the public, and a 45-day treatment regimen with doxycycline in the seeds was initiated. No further deaths of birds with hepatomegaly or splenomegaly were observed after the first day of treat

The prevalence of antibodies to reovirus type 3 in adults with idiopathic cholestatic liver disease.

Recent evidence suggests that neonatal infection with Reovirus type 3 (Reo-3) may play an important role in the pathogenesis of certain idiopathic cholestatic liver diseases of children. Whether this virus is of pathogenetic importance in similar diseases seen in adults is unclear. In this study, sera from 22 adults with idiopathic cholestatic liver disease (16 primary biliary cirrhosis (PBC) and six primary sclerosing cholangitis (PSC] were tested for antibody to Reo-3 virus by a standard hemagglutination inhibition assay. The results were compared to antibody prevalence in ten adults with va

Virus detection by short read high throughput sequencing in a high virus low cellular background.

The safety of all biological products includes demonstrating the absence of adventitious viruses by testing various types of samples at different stages of the manufacturing process. Seven laboratories evaluated short-read high-throughput sequencing (HTS) for sensitivity and breadth of adventitious virus detection using viruses with distinct physicochemical and genome properties. These five viruses are currently designated as CBER NGS Virus Reagents and include: Epstein-Barr virus (EBV; or human herpes virus 4), feline leukemia virus (FeLV), respiratory syncytial virus (RSV), mammalian orthore

Environmental DNA/RNA for non-invasive early detection and monitoring of pathogen dynamics in Atlantic salmon (Salmo salar) recirculating aquaculture systems (RAS).

Recirculating Aquaculture Systems (RAS) provide efficient fish production with reduced water use and precise environmental control. However, their closed design facilitates susceptibility to pathogen persistence and disease outbreaks, highlighting the need for effective monitoring and treatments. Over a 12-week study, water samples (purified eDNA/eRNA) were evaluated in two freshwater RAS units at a commercial pre-smolt Atlantic salmon (Salmo salar) farm as a non-lethal and non-invasive alternative to tissue samples for early pathogen detection. A key aim was furthermore to elucidate the infec

First field evidence of aerosolised SGPV, ISAV-HPR0, and IPNV in Atlantic salmon RAS highlights transmission and biosecurity risks.

Recirculating aquaculture systems (RAS) for Atlantic salmon (Salmo salar L.) are closed-containment systems (CCS) offering biosecure, water-efficient farming conditions, yet pathogen transmission remains a critical concern. While horizontal transmission in water is well-documented in RAS, the potential for aerosol-mediated transmission remains underexplored. The current study was conducted at two commercial Faroese Atlantic salmon RAS smolt farms. At Smolt farm 1, aerosolised pathogens were monitored using two aerosol samplers (Coriolis+ and Coriolis Compact, Bertin Technologies SAS, France),

Viral Heterogeneity in an Avian Reovirus Isolate Despite Plaque Purification and Differences in Segment-Wise Abundance Between Different Amplification and Purification Steps.

Avian reovirus (ARV) remains a significant concern in the poultry industry. Three rounds of plaque purification are routinely employed to obtain clonal populations of a viral isolate for experimental purposes. However, the effectiveness of this approach in achieving viral homogeneity has usually been taken for granted, especially in older publications, when there was no means of checking the purity. This study aimed to determine the purity of a plaque-purified ARV isolate (strain AL) using conventional PCR, Sanger sequencing, and whole-genome sequencing (WGS). WGS analysis of multiple samples

The Antibiotic Neomycin Enhances Coxsackievirus Plaque Formation.

Coxsackievirus typically infects humans via the gastrointestinal tract, which has a large number of microorganisms collectively referred to as the microbiota. To study how the intestinal microbiota influences enteric virus infection, several groups have used an antibiotic regimen in mice to deplete bacteria. These studies have shown that bacteria promote infection with several enteric viruses. However, very little is known about whether antibiotics influence viruses in a microbiota-independent manner. In this study, we sought to determine the effects of antibiotics on coxsackievirus B3 (CVB3)

Optimized polypeptide for a subunit vaccine against avian reovirus.

Avian reovirus (ARV) is a disease-causing agent. The disease is prevented by vaccination with a genotype-specific vaccine while many variants of ARV exist in the field worldwide. Production of new attenuated vaccines is a long-term process and in the case of fast-mutating viruses, an impractical one. In the era of molecular biology, vaccines may be produced by using only the relevant protein for induction of neutralizing antibodies, enabling fast adjustment to the emergence of new genetic strains. Sigma C (SC) protein of ARV is a homotrimer that facilitates host-cell attachment and induce the

Reovirus delays diabetes onset but does not prevent insulitis in nonobese diabetic mice.

Mice infected with reovirus develop abnormalities in glucose homeostasis. Reovirus strain type 3 Abney (T3A) was capable of systemic infection of nonobese diabetic (NOD) mice, an experimental model of autoimmune diabetes. Reovirus antigen was detected in pancreatic islets of T3A-infected mice, and primary cultures of pancreatic islets from NOD mice supported T3A growth. Significantly fewer T3A-infected animals compared to uninfected controls developed diabetes. However, despite the alteration in diabetes penetrance, insulitis was evident in T3A-infected mice. These results suggest that viral i

Novel goose parvovirus as a major risk factor for red skin and bristle feather syndrome in meat ducks: Epidemiological associations with duck circovirus and reovirus.

In recent years, a disease characterized by dorsal skin reddening and feather development disorder, termed "red skin and bristle feather syndrome" (RSBFS), has emerged in meat duck farms in China, resulting in substantial economic losses. To clarify the major etiological risk factor, their epidemiological patterns, and molecular characteristics, we collected 300 clinical samples from symptomatic and asymptomatic ducks across eight major duck-producing provinces in China between February and July 2025. Multipathogen screening was conducted using quantitative real-time PCR, followed by phylogene

A new real-time RT-PCR for the detection of piscine orthoreoviruses PRV-1 and PRV-3.

Piscine orthoreoviruses (PRV) are often associated with pathologies affecting various salmonid species, especially farmed species. Both surveillance and research purposes require diagnostic molecular-based tools able to detect the most frequent genotypes, namely PRV-1 and PRV-3. A new one-step reverse-transcription real-time PCR (RT-qPCR) was set up targeting a conserved region of the L1 segment. Based on the French standard NF U47-600, this PCR method proved to be specific to PRV, excluding other salmonid viruses. The assay can detect 1000 copies of a synthetic transcript of PRV-3, pure or mi
