Eight new ent-pimarane diterpenoids (1-8), along with three known compounds (9-11), were isolated from the aerial parts of Sigesbeckia pubescens. The structures of the new ent-pimarane diterpenoids were established by extensive spectroscopic techniques, X-ray diffraction crystallography, ECD calculations and Mo2(OAc)4-induced ECD. The isolated compounds were evaluated for their myocardial protective activities in an H9c2 cell hypoxia/reoxygenation (H/R) induced myocardial ischemia-reperfusion injury model. Bioassay results showed that pubescens B (2), pubescens H (8) and darutigenol (9) enhanc

Cercospora leaf blight (CLB) is the most devastating foliar disease of soybean in the US Gulf South, causing significant economic impacts throughout the growing region. The sole causal agent of CLB was thought to be Cercospora kikuchii, but we now recognize that the disease is predominantly caused by Cercospora cf. flagellaris and Cercospora cf. sigesbeckiae in North America. However, a lack of information about the spatial population dynamics of the causal agents and the disease cycle, including the most important sources of inoculum, hinders the development of new and effective management to

Macrophages-mediated inflammation is involved in the regulation of rheumatoid arthritis (RA). Sigesbeckiae Herba (SH) has been traditionally used for rheumatism. However, the bioactive ingredients of SH are still unclear. Recently, we isolated a compound (Leocarpinolide B, LB) from SH and identified its potent anti-inflammatory and antioxidant effects on RAW264.7 macrophages for the first time. LB effectively inhibited excessive production of nitric oxide (NO), prostaglandin E2 (PGE2), cytokines (IL-6, TNF-α and MCP-1), and the expression of cyclooxygenase-2 (COX-2) and inducible nitric oxide

Six novel diterpenoids (1-6) and seventeen known compounds (7-23) were isolated from the aerial parts of Siegesbeckia pubescens Makino. The chemical structures of the new compounds were established through comprehensive spectroscopic characterization, including HR-ESI-MS and 1D/2D NMR analyses, and their absolute configurations were determined by ECD calculations. The anti-inflammatory effect of all compounds against lipopolysaccharide (LPS)-induced RAW 264.7 cells was tested. The results indicated that compounds 2-18 and 20-23 significantly suppressed nitric oxide (NO) production.

Rheumatoid arthritis (RA) is a chronic autoimmune disease triggered by a cascading inflammatory response. Sigesbeckia Herba (SH) has long been utilized as a traditional remedy to alleviate symptoms associated with rheumatism. Our previous study found that leocarpinolide B (LB), a sesquiterpene lactone isolated from the whole plant of SH, possesses potent a anti-inflammatory effect on macrophages. This study was designed to evaluate the therapeutic effects of LB on RA, and further investigate the underlying mechanisms. In collagen type II-induced arthritic mice, LB was demonstrated to decrease

BACKGROUND: Ischemic stroke is a life-threatening cerebrovascular disease with limited therapeutic options. During the progression of acute ischemic stroke (AIS), neutrophil-involved inflammation mediated by high mobility group box 1 (HMGB1) considerably contributes to intensification of neuronal injury. Siegesbeckia orientalis L. (SO), one of the primary sources of Sigesbeckiae Herba, is promising in anti-neuroinflammation and neutrophil function modulation. Consequently, it is supposed that SO could fight against neuronal inflammatory injury following AIS.
PURPOSE: The current study struggl
