Research progress of Sinomenium in the treatment of rheumatoid arthritis and suggestions for future research.

Rheumatoid arthritis (RA) is a chronic autoimmune joint disease. Its main pathological manifestations are joint cartilage, bone tissue injury, synovial hyperplasia, and chronic inflammation. At present, the pathogenesis of the disease has not been fully defined, and delaying the disease to improve joint function is the existing treatment. Sinomenium (SIN) is an effective ingredient in Sinomenium acutum (Thunb.) Rehd. et Wils., with anti-inflammatory, analgesic, immunosuppressive, anti-tumor, and other pharmacological activities, which is quite effective in the treatment of RA. There are many S

Functional Characterization of Two Methyltransferases Involved in Benzylisoquinoline Alkaloid Biosynthesis in Sinomenium acutum.

Benzylisoquinoline alkaloids (BIAs) are a diverse group of plant secondary metabolites with significant pharmacological activities, where methylation reactions catalyzed by methyltransferases play crucial roles in structural diversification and bioactivity modulation. In this study, we identified and functionally characterized two key methyltransferases, Sinomenium acutum 6-O-methyltransferase (Sa6OMT) and S. acutum coclaurine N-methyltransferase (SaCNMT), involved in BIA biosynthesis in S. acutum, a traditional Chinese medicinal plant with anti-rheumatoid arthritis properties. Candidate genes

Iterative enrichment cultivation and multiomic analysis reveal potential endophytic bacteria affecting the sinomenine synthesis in Sinomenium acutum.

Endophytes play important roles in plant metabolite synthesis, and certain strains were capable of producing bioactive compounds identical to those of their hosts. However, it remains unknown whether culturable endophytes of S. acutum can synthesize intermediate metabolites for the plant principal bioactive compound-sinomenine (SIN) or the compound itself. In this study, we investigated the successions of the culturable bacterial community and the alkaloid profiles within S. acutum endophytes across ten iterative enrichment cultivations using Czapek-Dox and Gause's No. 1 chemically defined med

Sinomenine promotes flap survival by upregulating eNOS and eNOS-mediated autophagy via PI3K/AKT pathway.

Large skin defects and surgical tissue reconstructions are frequently covered utilizing random flaps. The flap has the advantage of being designed according to the size and shape of a surgical wound. However, the necrosis of the distal part of the flap restricts the clinical application of flaps. Sinomenine (SIN) is the major active component of sinomenium acutum. SIN has been demonstrated to inhibit oxidative stress and stimulate autophagy in a cell, animal, and clinical studies. The protective and proliferative effects of sinomenium on HUVECs were evaluated by scratched test, CCK-8, and EDU

[Pharmacological Mechanisms of Boiogito and Bofutsushosan in Diabetes and Obesity Models].

The antihyperglycemic activities of extracts of boiogito (BOT) and bofutsushosan (BTS) were investigated in streptozotocin-induced (STZ)-diabetic mice. BOT extract containing Stephania tetrandra S. MOORE root (stephania), has more potent antihyperglycemic activity than BOT extract containing sinomenium stem (sinomenium). Extracts of stephania and astragalus root (astragalus) exert combined effects in the antihyperglycemic and insulinotropic activities of BOT extract. Fangchinoline, but not tetrandrine, in stephania plays a role in its activity. Formononetin in astragalus potentiates the action
