Orthomarburgviruses are the etiological agents of human Marburg virus disease, with case fatality rates reaching up to 90%. Of the limited known targets for protective antibodies on the orthomarburgvirus surface glycoprotein (GP), the GP2 wing region is undefined structurally, and its recognition by antibodies is poorly understood. We report a comprehensive genetic and structural analysis of four independent broadly reactive antibody lineages that target a common epitope within the GP2 wing, including macaque antibodies CM10, CM11.1, and CM12.1 and murine antibody 30G4. Co-crystal structures r

SUMMARYMarburg virus disease (MVD) is a highly fatal illness closely resembling Ebola disease, caused by either Marburg virus (MARV) or Ravn virus (RAVV). It typically begins with a sudden onset of high fever, headache, and malaise, followed by severe gastrointestinal symptoms, bleeding disorders, and, in the most severe cases, multi-organ failure. Case fatality rates vary widely, from 24% up to 90%. MARV and RAVV are zoonotic pathogens, with fruit bats recognized as the main reservoir. Human-to-human transmission generally occurs through direct exposure to infected bodily fluids. Historically

In November 2025, Marburg virus caused the first outbreak of Marburg virus disease (MVD) in Ethiopia. By December 15, a total of 14 laboratory-confirmed cases were reported, including 9 deaths, corresponding to a case fatality ratio of 64.3% among confirmed cases. In the absence of licenced vaccines or antivirals, non-pharmaceutical interventions (NPIs) were implemented to control transmission. We developed a stochastic epidemic model incorporating a stochastic exponential growth process with reporting adjustment to assess the effectiveness of NPIs. The reporting ratio was modelled using a bet

Marburg virus (MARV), a highly pathogenic member of the Filoviridae family, causes severe hemorrhagic fever with a high case fatality rate and currently lacks effective therapeutics. The viral entry process, mediated by the interaction between the MARV glycoprotein (GP) and host receptor C-type lectin domain family 4 member M (CLEC4M) (L-SIGN), represents a critical target for early-stage intervention. The active compounds from BindingDB and the decoy from DUDE were used. The RDKit was used for feature engineering. Machine learning models were trained on an initial dataset consisting of 56 act

After the 2023 Equatorial Guinea Marburg virus (MARV) outbreak, surveillance of 181 persons in southern Cameroon detected MARV antibodies in 3 persons and Ebola virus antibodies in 7. Testing of 289 captured bats, including 158 Rousettus aegyptiacus bats, did not detect MARV RNA. Enhanced surveillance for regional filovirus spillover risks is warranted.

INTRODUCTION: Marburg virus disease (MVD) occurs in sporadic and unpredictable outbreaks, making conventional human efficacy studies of vaccines and therapeutics impractical. This review summarizes animal models of MVD and their utility for pathogenesis research and evaluation of vaccines and therapeutics.
AREAS COVERED: Herein, the authors review rodent, ferret, and nonhuman primate models of MVD infection, focusing on how each reflects human disease and supports different stages of countermeasure development. Furthermore, the authors discuss their virological and pathological features and m

The recent outbreaks of Marburg virus disease (MVD) in Tanzania and Sudan virus disease (SVD) in Uganda reveal the growing threat of filoviruses in East Africa. These highly lethal viruses, with case fatality rates exceeding 50%, continue to challenge regional health systems due to delayed detection, weak surveillance networks, and limited cross-border coordination. The outbreak in Tanzania's Kagera region, a major transit hub, highlights the risk of international spread, while Uganda's struggle with Sudan virus further emphasizes the urgent need for regional infectious disease surveillance. The emergence of significant viral pathogens and their escalating threats to human and animal health have intensified scientific interest in bats as disease vectors and natural reservoir hosts. Among chiropteran species, the Egyptian rousette (Rousettus aegyptiacus) assumes particular epidemiological significance as the established natural reservoir for orthomarburgviruses, encompassing the two recognized members Marburg virus and Ravn virus. While contemporary research predominantly emphasizes the detection and characterization of specific zoonotic agents within these populations, fundamental

Ebola virus disease and Marburg virus disease are severe, highly lethal infections caused by filoviruses, characterized by rapid transmission, complex clinical management, and major socio-economic impact. A narrative review of recent outbreaks between 2021 and 2025 underscores persistent systemic gaps that compromise early detection and effective containment. The recurrence of Ebola and Marburg outbreaks demonstrates that, although progress has been made since the 2014-2016 West African epidemic, substantial vulnerabilities persist.

Marburg virus disease (MVD) is re-emerging across Africa as a high-consequence zoonosis shaped by expanding ecological suitability, repeated spillover, and uneven surveillance capacity. This review synthesizes current evidence on the ecological, epidemiological, and operational determinants of contemporary Marburg virus (MARV) emergence. We conceptualize MVD as an ecological-emergence system produced by interactions among reservoir-host biology, environmental change, human exposure, health-system readiness, and mobility, rather than as a series of isolated outbreaks. Recent detections in multi

INTRODUCTION: Marburg virus (MARV) causes a potentially highly lethal haemorrhagic fever disease that threatens global health security, and knowledge of viral persistence, long-term clinical outcomes among recovered persons and protective immunity remain limited. Existing protocols and technical guidelines provide a framework for safe sample collection, clinical monitoring and outbreak response, yet systematic approaches to investigate these aspects in African settings are scarce. This protocol describes the Partnership Research on MARV (PREMAV) following the 2024 outbreak in Rwanda. PREMAV is

BACKGROUND: Magnetic resonance cholangiopancreatography (MRCP) is useful to assess exocrine pancreatic function by combining rapid imaging acquisition with the administration of secretin, a gastrointestinal hormone that stimulates the secretion of bile and pancreatic juice. However, extensive data on this method are lacking. This study aimed to determine whether MRCP with secretin administration is able to simultaneously detect alterations of both the pancreatic ducts and exocrine pancreatic function.
METHODS: All subjects older than 18 years who underwent magnetic resonance imaging (MRI) and
