PURPOSE OF REVIEW: Atypical chemokine receptor-1 (ACKR1)/Duffy antigen receptor of chemokines (DARC)-associated neutropenia (ADAN; OMIM 611862), previously named benign ethnic neutropenia, and present in two-thirds of individuals identifying as Black in the USA, is associated with mild to moderate decreases in peripheral neutrophil counts that nevertheless do not lead to increased infections. Consequently, recent initiatives have sought to establish normal neutrophil count reference ranges for ADAN, considering it a normal variant rather than a clinical disorder requiring medical intervention.

OBJECTIVE: Blood group antigen frequencies vary across populations and influence transfusion compatibility and alloimmunization risk; data on minor systems in northern Saudi Arabia are limited. This study determined Kidd (Jk), Lutheran (Lu), and Duffy (Fy) antigen and phenotype frequencies among Al-Qurayyat blood donors and compared them with other populations.
METHODS: In this cross-sectional study, 240 healthy blood donors were serologically typed for Kidd (Jka, Jkb), Lutheran (Lua, Lub), and Duffy (Fya, Fyb) antigens. Using the chi-square test, Phenotype frequencies were compared with publ

Chemokines represent a family of potent biological mediators. Within the group of receptors mediating their effects, a promiscuous receptor has been found which is able to bind and inactivate diverse chemokines of both C-C and C-X-C families. It is co-localized with blood group antigens of the Duffy system on the same glycoprotein and expressed on red blood cells as well as post-capillary blood vessels. In the present study three aspects of Duffy pathophysiology were studied: firstly the amount of IL-8 and RANTES binding to red blood cells and its correlation to disease activity of psoriatic p

BACKGROUND: Sickle cell disease (SCD) patients undergo multiple red blood cell (RBC) transfusions and are regularly exposed to low-prevalence (LP) antigens specific to individuals of African descent. This study evaluated the prevalence of antibodies against LP antigens in SCD patients and the need to identify these antibodies in everyday practice.
STUDY DESIGN AND METHODS: Plasma from 211 SCD patients was tested with RBCs expressing the following LP antigens: RH10 (V), RH20 (VS), RH23 (DW ), RH30 (Goa ), KEL6 (Jsa ), and MNS6 (He).
RESULTS: Nine LP antibodies were found in eight patients (3.

BACKGROUND AND OBJECTIVES: Serological classification of RhD-negative and weak D is often insufficient in admixed populations, where diverse RHD variants have clinical implications. In Brazil's highly admixed population, RhD interpretation and donor-recipient matching are particularly challenging. We used Duffy phenotyping to support ancestry inference and contextualize Rh variants. Understanding RHD alleles and their associations with RhCE and Duffy is essential for transfusion safety. Here, we characterize RHD variants in Brazilian donors and relate them to RhCE and Duffy phenotypes across r

Various models have been developed to study red blood cell (RBC) alloimmunization. While studies using these models have primarily examined general features of this response, identifying key characteristics of antigen-specific B cells is critical when seeking to define regulators of alloimmune responses. Unlike analysis of antigen-specific T cells, which requires peptide loaded in the appropriate major histocompatibility class structure, B cells recognize native antigens, which requires the generation of tools that incorporate individual antigens into multimeric constructs to enhance B cell re

BACKGROUND: Duffy glycoprotein, belonging to blood group alloantigens, is also recognized as chemokine-binding protein, therefore the role of Duffy antigen in chemokine distribution has been postulated. Duffy positive phenotype prevalent among Caucasians is associated with antigen expression on erythrocytes and endothelium of several organs including kidney. The role of anti-Duffy antibodies significant in transfusiology may be also important in kidney transplantation.
CASE REPORT: The case of renal transplant recipient with Fy (a-b+) phenotype, possessing anti Fya antibodies, with unfortunat

The assignment of blood group loci proceeds more slowly than that of biochemical markers, since it depends largely on pedigree analysis and these studies take longer to achieve significance than some of the rapid modern methods. However, seven structural blood group loci ABO, MNSs, Rh, Fy, Sc, Xg, and Ch--Rg are confidently assigned, and four more provisionally or tentatively sited. Other loci are recognized as part of linkage groups that will help in the eventual assignment to their chromosomes. There is little linkage information, and no assignments, for other independent loci involved in th

BACKGROUND AND OBJECTIVES: A lateral flow assay for simultaneous blood group typing of ABO, RhD, C, E, c, e, Cw and K with stable end-point and without centrifugation is in routine use since several years (MDmulticard® ). The typing of extended phenotype parameters belonging to the Duffy, Kidd, MNSs blood group systems and others, however, has not yet been demonstrated for this technique. Reliable detection of Fyx , a weak Fyb phenotype with a pronounced quantitative reduction of the number of Fyb antigens on the erythrocyte surface, remains a weakness of current serological blood grouping tec

Duffy blood group negative erythrocytes from blacks are refractory to invasion by Plasmodium knowlesi merozoites in vitro, and blacks with this genotype are resistant to infection by P. vivax in vivo. In order to evaluate in a direct manner the role of Duffy blood group determinants in invasion by P. knowlesi merozoites, we studied erythrocytes from three rare non-black Duffy negative individuals, Fy(a-b-), in whom the Duffy negative phenotype probably represents a mutation and not the introduction of the black Fy gene. These cells were resistant to invasion by P. knowlesi in vitro indicating

The transfusion histories over a 33-month period of 50 patients with sickle cell disease were reviewed to determine the frequency of alloimmunization to red cell antigens following transfusion in these patients. There were 30 females and 20 males, aged 19--49 years. Eighteen (36%) were immunized of which thirteen were females. Five of the patients have formed only one antibody so far, while the other 13 have formed two or more. Thirty-six antibodies were identified: 16 against various Rh antigens, 12 anti-Lewis, 5 anti-Kell and one each of anti-Jka, -Fya and -M. The immunized patients received

Sickle cell disease (SCD) is associated with a pro-inflammatory state, characterized by an elevated baseline leukocyte count and inflammatory cytokines. Inflammation, white blood cell (WBC) adhesion to vascular endothelium with subsequent endothelial injury, and repeated ischemia-reperfusion injury contribute to disease pathogenesis. Identification of genetic polymorphisms that may modulate disease severity in SCD is becoming a field of interest. The Duffy blood group antigen has been identified as a receptor for various chemokines involved in neutrophil activation and trafficking. This study

Malaria has exhibited the strongest known selective pressure on the human genome in recent history and is the evolutionary driving force behind genetic conditions, such as sickle-cell disease, glucose-6-phosphatase deficiency, and some other erythrocyte defects. Genomic studies (e.g., The 1000 Genomes project) have provided an invaluable baseline for human genetics, but with an estimated two thousand ethno-linguistic groups thought to exist across the African continent, our understanding of the genetic differences between indigenous populations and their implications on disease is still limite

Most discussions of blood transfusion cover risks and side effects, while the daily benefits of donor systems and transfusion concepts for modern medicine are taken for granted. Even though in this report side effects are a major part of the content, we should be aware that today's component concept leads to a remarkable quality level of blood products. After a historical introduction we present a patient with transfusion transmitted anti-hepatitis A IgM antibodies and a delayed type hemolytic transfusion reaction induced by a boostered anti-Fyb alloantibody. This case demonstrates that the sa

After several transfusions with D-negative blood, an O Rh-negative women was apparently sensitized to the C and D antigens. In her prenatal workup, it became evident that she had in fact not been sensitized to D but to G, which initially appeared as anti-D plus anti-C. This sensitization pattern is an unusual occurrence in itself. Moreover, the fetus was affected significantly and was delivered at 32 1/2 weeks with moderate hemolytic disease of the newborn. Consideration is given to points regarding current methods of screening Rh-negative women for transfusion, the use of anti-Rh immune globu

The antigens of the Duffy blood group system (DARC) act as a receptor for the interleukin IL-8. IL-8 plays an important role in the pathogenesis of chronic periodontitis due to its chemotactic properties on neutrophils. The aim of this study was to investigate a possible association of Duffy blood group gene polymorphisms with the -353T>A, -845T>C and -738T>A SNPs of the IL8 gene in chronic periodontitis. One hundred and twenty-four individuals with chronic periodontitis and 187 controls were enrolled. DNA was extracted using the salting-out method. The Duffy genotypes and IL8 gene promoter po

Dysregulated inflammation in response to inciting events leads to severe end-organ dysfunction and failure, contributing to long-term morbidity and, ultimately, potentially death. Duffy antigen receptor for chemokines (DARC), a major regulator of inflammation, plays key roles in a variety of acute and chronic inflammatory states. DARC minimizes the end-organ injury induced in response to an inciting event by acting as a "sink" to soak up excessive inflammatory cytokines and chemokines. Data demonstrate how the lack of DARC functioning is associated with altered immunopathological and clinical

A man and his twin sister were found to have two types of circulating red cells. The man had 89% A1 cells of his own and 11% A1B cells derived from his sister. The woman had 12% A1B cells of her own and 88% A1 cells derived from her brother. The two lines of red cells also had different Duffy and Kidd groups. The lymphocyte karyotype in the brother was 68% XY and 32% XX, and that in the sister was 60% XX and 40% XY. The possible mechanisms for producing different proportions of red cells and lymphocytes are discussed.
