Generation of human iPSC line from a patient with Tetralogy of Fallot, YAHKMUi001-A, carrying a mutation in TBX1 gene.

The human induced pluripotent stem cell (iPSC) line YAHKMUi001-A was derived from the dermal fibroblasts of a patient with Tetralogy of Fallot (TOF), with a mutation in the TBX1 gene (c.928G > A). The skin fibroblasts were obtained from a 4-year-old boy, and were infected with Sendai virus expressing the Yamanaka factors. The YAHKMUi001-A iPSC line expresses pluripotent stem cell markers, displays a normal karyotype, and has the capacity to differentiate into 3 germ layers. This cell line model can be a good tool to study the pathological mechanism of the TBX1 gene mutations associated with TO

Generation of integration-free induced pluripotent stem cells (GZHMUi001-A) by reprogramming peripheral blood mononuclear cells from a 47, XXX syndrome patient.

47, XXX syndrome is one of several sex-chromosomal aneuploidies, and it has an incidence of approximately 1/1000 in newborn females. Because of heterogeneity in X-inactivation, these patients may exhibit a variety of clinical symptoms. Here, we report the generation of an integration-free human induced pluripotent stem cell line (GZHMUi001-A) by using Sendai virus to reprogram peripheral blood mononuclear cells from a 47, XXX syndrome patient with premature ovarian failure. This 47, XXX iPS cell line has characteristics of pluripotent stem cells and is a useful tool for the investigation of th

Viral antibodies in multiple sclerosis.

The sera of 176 MS patients and of 150 healthy adult controls were assayed for antibodies against mumps, rubella, Sendai and herpes simplex viruses, a higher prevalence of measles c.f.a. having already been demonstrated in the MS patients. The CSF of 48 of the MS patients were subjected to the same tests. The patients differed from the controls in a higher prevalence of h.i.a. to mumps and of c.f.a. to herpes simplex. For the latter, but not for the former, the prevalence was statistically higher only in patients treated with immunosuppressants. To date measles seems to be the most seriously i

Generation of a human induced pluripotent stem cell line (ZZUNEUi040-A) from a male patient with hypertrophic cardiomyopathy carrying a heterozygous TPM1 (c.644C > T) mutation.

Tropomyosin 1 (TPM1) encodes tropomyosin alpha-1 chain, a key cardiac sarcomere component, while TPM1 mutations cause hypertrophic cardiomyopathy (HCM), a major cause of sudden cardiac death in the young. We generated the human iPSC line ZZUNEUi040-A from peripheral blood mononuclear cells of a 10-year-old male with non-obstructive asymmetric HCM carrying a heterozygous TPM1 c.644C > T (p.Ser215Leu) mutation via Sendai virus non-integrating reprogramming technology. The line showed normal karyotype (46,XY), undifferentiated hPSC-State markers, and trilineage differentiation potential, serving

Protocol for the induction of human spinal motor neurons from human induced pluripotent stem cells for studying amyotrophic lateral sclerosis.

Here, we present a protocol for inducing spinal lower motor neurons (LMNs) from human induced pluripotent stem cells (iPSCs). We describe steps for preparation of a chemically induced transitional state (CTraS), transduction with Sendai virus, and LMN differentiation and maintenance. We then detail procedures for live imaging for single-cell-based survival analysis and neurite length of LMNs using BioStation and immunocytochemistry for induction efficiency check. This protocol is optimized for amyotrophic lateral sclerosis (ALS) research and large-scale screening. For complete details on the u

Generation and characterization of human iPSC line SANi011-A from a patient with an inherited platelet disorder carrying the heterozygous FLI1 c.297del variant.

FLI1, a member of the ETS transcription factor family, is associated with Paris-Trousseau thrombocytopenia, and germline FLI1 mutations have been identified in patients with inherited platelet disorders. We generated the iPSC line SANI011-A from a patient carrying a de novo heterozygous nonsense mutation, FLI1 c.297del. Proerythroblasts derived from the patient's peripheral blood were reprogrammed into iPSCs using the non-integrative Sendai virus (SeV) delivery method. The resulting iPSC line exhibited normal karyotype, expressed pluripotent markers, and demonstrated the capacity for trilineag

Generation of an induced pluripotent stem cell line from a Wilson's disease patient carrying both c.2333G > T and c.2621C > T mutations.

Wilson's disease (WD) is an autosomal recessive genetic disorder caused by the mutation of ATP7B gene encoding ATP7B protein (copper ion transporter ATPase β peptide). Functional impairments of ATP7B protein causecopper transport disorders in liver cells, eventually resulting in abnormal accumulation of copperand pathological manifestations. In this study, human renal epithelial cells obtained from a patient with WD were reprogrammed by a non-integrated Sendai virus to generate a patient-specific induced pluripotent stem cell (iPSC) line. The iPSC line expressed pluripotency markers, showed nor

Generation of human induced pluripotent stem cell lines derived from four Rett syndrome patients with MECP2 mutations.

Rett syndrome is characterized by severe global developmental impairments with autistic features and loss of purposeful hand skills. Here we show that human induced pluripotent stem cell (hiPSC) lines derived from four Japanese female patients with Rett syndrome are generated from peripheral blood mononuclear cells using Sendai virus vectors. The generated hiPSC lines showed self-renewal and pluripotency and carried heterozygous frameshift, missense, or nonsense mutations in the MECP2 gene. Since the molecular pathogenesis caused by MECP2 dysfunction remains unclear, these cell resources are u

Generation of induced pluripotent stem cell lines from a sister pair who suffered post-partum or recurrent Spontaneous Coronary Artery Dissections.

Spontaneous Coronary Artery Dissection (SCAD) results from a bleed within a coronary artery wall that impairs blood flow as it expands. It is the most common cause of myocardial infarction in pregnant women. Here, peripheral blood mononuclear cells from two sisters who had suffered SCADs were reprogrammed using Sendai Virus. Expression of pluripotency markers, capability to differentiate to the three germ layers, and cellular integrity were confirmed. This is the first report of a SCAD family induced pluripotent stem cell (iPSC) cohort, including a sister who suffered post-partum SCAD, and one

Generation of DMBi002-A human induced pluripotent stem cell line from patient with Spinal muscular atrophy type 3.

Spinal Muscular Atrophy (SMA) is a genetic neuromuscular disease caused by mutations inSMN1 gene encoding survival motor neuron (SMN) protein. Lack of this protein leads to progressive loss of motor neurons and therefore to gradual loss of signal transmission between motor neurons and skeletal muscle cells. As a consequence, patients develop muscle atrophy and lose the ability to move independently, what is also related to problems with breathing and swallowing. Here, we describe the generation of human induced pluripotent stem cells (hiPSC) from peripheral blood mononuclear cells (PBMC) of ad
